Establishment and Comprehensive Molecular Characterization of an Immortalized Glioblastoma Cell Line from a Brazilian Patient
Glioblastoma (GBM) is the most common and aggressive primary brain tumor in adults, with few effective treatment strategies. The research on the development of new treatments is often constrained by the limitations of preclinical models, which fail to accurately replicate the disease’s essential characteristics. Herein, we describe the obtention, molecular, and functional characterization of the GBM33 cell line. This cell line belongs to the GBM class according to the World Health Organization 2021 Classification of Central Nervous System Tumors, identified by methylation profiling. GBM33 expresses the astrocytic marker GFAP, as well as markers of neuronal origin commonly expressed in GBM cells, such as βIII-tubulin and neurofilament. Functional assays demonstrated an increased growth rate when compared to the U87 commercial cell line and a similar sensitivity to temozolamide. GBM33 cells retained response to serum starvation, with reduced growth and diminished activation of the Akt signaling pathway. Unlike LN-18 and LN-229 commercial cell lines, GBM33 is able to produce primary cilia upon serum starvation. In summary, the successful establishment and comprehensive characterization of this GBM cell line provide researchers with invaluable tools for studying GBM biology, identifying novel therapeutic targets, and evaluating the efficacy of potential treatments.
Utilization of Moringa oleifera seeds as a biosorbent for diclofenac removal in the contaminated aquatic systems
During recent years, pharmaceuticals and their metabolites have been increasingly found in water bodies and diclofenac is one of the pharmaceuticals residues most frequently detected thus far. The aim of this work was to evaluate the Moringa oleifera seeds as an alternative for diclofenac (DCF) removal in water samples. The batch procedure for DCF removal at the optimized conditions (25 mL of 10.0 mg L−1 DCF, pH 5.0, extraction time of 30 min and M. oleifera mass of 2.0 grams) achieved adsorption of 100% of DCF in real water samples. The pseudo-second-order kinetic described the adsorption rate-controlling step. The adsorption isotherms were fitted by Langmuir, Freundlich and Sips models and the sorption capacity of the biosorbent is spontaneously favourable. The maximum adsorptive capacity was estimated at 32.05 mg g−1 and 32.85 mg g−1 by Langmuir and Sips models, respectively. The advantages of this procedure include good reproducibility in the removal of DCF even at low concentrations in real samples and does not require an additional step of pre-treatment of the adsorbent. The results highlight the potential of M. oleifera seeds as a cheap, environmentally friendly alternative for the removal of DCF from polluted water.
Saúde digital e a plataformização do Estado brasileiro
A implementação da saúde digital constitui um enorme desafio para a Saúde Coletiva, sendo urgente abrir o debate sobre os impactos mais imediatos das tecnologias digitais nas políticas de saúde. A saúde digital compreende a incorporação de novas tecnologias e potencialmente reconfigura relação entre Estado e sociedade, em um processo denominado plataformização - de gestão dos serviços de saúde por meio da interpretação de grandes volumes de dados. Este trabalho traça um panorama histórico sobre as políticas brasileiras de informação e analisa a saúde digital como um caso de plataformização do Estado Brasileiro. Para tanto, analisa a estratégia brasileira de saúde digital partir de três dimensões: a concentração de dados, os usuários-consumidores e a privatização das infraestruturas públicas. Por fim, busca tornar nítida a tendência global a favor de uma inovação que escamoteia a expectativa pela digitalização como dinamizadora da reprodução capitalista.
Dimensões subjetivas na Saúde Digital
The heterogeneous and contradictory structures of global capitalism have been shaping the social meanings about Digital Health in many ways and, thereby, its development and regulation as well. For a better understanding of these issues, in this literature review we highlight that these structures include certain subjective dimensions that must be investigated in more detail. For this purpose, we analyze how Digital Health becomes a space both for the expansion of human potential and for the intensification of these structural contradictions. We discuss how the modes of capitalist governmentality would be updated along with these movements, subjectively shaping pathologies, patients and health care. We analyze how these processes can unfold into behavioral techniques that can be embedded in health and wellness devices and applications, as well as their consequences and potential risks, which requires regulatory attention along with social participation As heterogêneas e contraditórias estruturas do capitalismo global vêm moldando de diferentes formas os significados sociais acerca da Saúde Digital e, com isso, também seu desenvolvimento e regulação. Para uma melhor compreensão destas questões, nesta revisão de literatura destacamos que tais estruturas contêm certas dimensões subjetivas que precisam ser investigadas de forma mais detida. Para isso, analisamos como a Saúde Digital se torna um espaço tanto de expansão das potencialidades humanas quanto para o acirramento dessas contradições estruturais. Discutimos como os modos de governamentalidade capitalista seriam atualizados junto a esses movimentos, moldando subjetivamente patologias, pacientes e o cuidado em saúde. Analisamos como esses processos podem se desdobrar em técnicas comportamentais que podem ser embarcadas em dispositivos e aplicativos de saúde e bem-estar, bem como suas consequências e riscos potenciais, o que exige atenção regulatória com participação social
Synthesis and Cation Exchange of LTA Zeolites Synthesized from Different Silicon Sources Applied in CO2 Adsorption
Zeolites have a well-ordered crystalline network with pores controlled in the synthesis process. Their composition comprises silicon and aluminum, so industrial residues with this composition can be used for the synthesis of zeolites. The use of zeolites for CO2 adsorption is feasible due to the characteristics that these materials have; in particular, zeolites with a low Si/Al ratio have greater gas adsorption capacities. In this work, the synthesis of LTA (Linde Type A) zeolites from silica fumes obtained from the industrial LIASA process and light coal ash is presented. We explore three different synthesis routes, where the synthesized materials undergo cation exchange and are applied in CO2 adsorption processes. Studying the synthesis processes, it is observed that all materials present characteristic diffractions for the LTA zeolite, as well as presenting specific areas between 6 and 19 m2/g and average pore distributions of 0.50 nm; however, the silica fume yielded better synthesis results, due to its lower impurity content compared to the light coal ash (which contains impurities such as quartz present in the zeolite). When applied for CO2 adsorption, the standard materials after cation exchange showed greater adsorption capacities, followed by the zeolites synthesized from silica fume and, finally, the zeolites synthesized from coal ash. By analyzing the selectivity of the materials for CO2/N2, it is observed that the materials in sodium form present greater selectivity when compared to the calcium-based materials.
Toward Better Outcomes in Audiology Distance Education: An Educational Data Mining Approach
This article introduces concepts and a general taxonomy used by the educational data mining (EDM) community, as well as examples of their applications, with the aims of providing audiology educators with a referential basis for developing this area. A narrative review was carried out to present an overview of EDM and its main methods. Some of these methods were exemplified with analysis of real data from an Internet-based specialization course on pediatric auditory rehabilitation. The review introduced EDM main concepts and applications and described methods from its area. Real data examples illustrated EDM use to predict interpersonal help-seeking, model interpersonal interaction, analyze students' trajectories within a course's module, and understand how students approached group assignments. Some of the insights provided by EDM to support teaching and learning processes were also described. EDM methods offer new tools to discover knowledge from digital traces (i.e., logs) and support key stakeholders (students, instructors, or course administrators) to raise awareness about course dynamics. This approach has the potential to foster a better understanding and management of educational processes in audiology distance education.
In Vitro Susceptibility to Metronidazole of Tritrichomonas foetus Bovine Isolates from Argentina
BackgroundTritrichomonas foetus is the etiologic agent of the sexually transmitted disease Bovine Trichomonosis (BT). In Argentina, BT is endemic and represents a relevant health problem that causes reproductive inefficiency in cattle and large economic losses. Metronidazole is the drug of choice in the treatment of BT. Treatment has been associated with a temporary resolution of the clinical signs but is not able to control the disease. In recent years, the apparition of in vivo and in vitro aerobic and anaerobic resistance leading to ineffective treatments has been reported.AimsThus, the aim of the present study was to explore the susceptibility of six different isolates of T. foetus under aerobic (AC) and anaerobic (ANC) conditions.Results and discussionSix isolates of T. foetus were obtained from samples of preputial smegma of bovine origin. Values of minimum lethal concentration and minimum inhibitory concentration were higher than those observed in other works and represent current data in Argentina and provide information to establish new treatment protocols.
Disseminated melanized fungal infection due to Cladosporium halotolerans in a dog coinfected with canine adenovirus-1 and canine parvovirus-2
This report presents the pathologic findings associated with disseminated infection due to Cladosporium halotolerans in a dog that was simultaneously infected with canine adenovirus-1 (CAdV-1) and canine parvovirus-2 (CPV-2). A 12-year-old, mixed breed dog, with a clinical history of neurological manifestations was submitted for routine autopsy due to poor prognosis. The principal pathologic findings were mycotic necrotizing nephritis, hepatitis, and splenitis with embolic dissemination to the brain resulting in mycotic necrotizing meningoencephalitis, ventriculitis, choroid plexitis, and obstructive hydrocephalus associated with intralesional and intravascular septate pigmented fungi. PCR and sequencing of the ITS region of fungi revealed that the intralesional fungal organisms had 82% nucleotide identity with members of the Cladosporium sphaerospermum complex of organisms. However, a PCR assay and sequencing of the beta tubulin gene confirmed that the organism identified in this dog had 100% nucleotide sequence identity with C. halotolerans . Using immunohistochemistry, intralesional antigens of CAdV-1 were identified within the epithelial cells of the liver and lungs; there was positive immunolabeling for CPV-2 antigens in degenerated cardiomyocytes. These findings confirmed the active participation of C. halotolerans in the development of disseminated cladosporiosis in this dog and represent a rare occurrence of concomitant infection with CAdV-1 and CPV-2.

Implementação de cifras caóticas sobre caos modular
Nesta dissertação, foi realizado um estudo aprofundado sobre Criptografia, Sistemas Caóticos, Cifras Caóticas, Caos Modular e um estudo final exaustivo do funcionamento da Cifra Caótica eLoBa. O objetivo desta dissertação foi implementar e melhorar a estrutura da Cifra Caótica eLoBa em outros sistemas caóticos além do Sistema Caótico de Lorenz, de modo a que, depois de implementados, foram recolhidas 1 milhão de chaves de cada Sistema Caótico para cada uma das duas sementes com apenas um caractere diferente. No final, são apresentados os resultados em tabelas e gráficos dos dados recolhidos da Bateria de Testes Estatísticos a Números Pseudo-aleatórios, NIST 800-22, com as amostras dos Sistemas Caóticos. De acordo com os dados recolhidos ficou confirmado que é possível a aplicação dos Sistemas Caóticos em Criptografia.
