Enzymatic Hydrolysis Systems Enhance the Efficiency and Biological Properties of Hydrolysates from Frozen Fish Processing Co-Products

Co-products from the frozen fish processing industry often lead to financial losses. Therefore, it is essential to transform these co-products into profitable goods. This study explores the production of fish protein hydrolysates (FPH) from three co-products: the heads and bones of black scabbardfish (Aphanopus carbo), the carcasses of gilthead seabream (Sparus aurata), and the trimmings of Nile perch (Lates niloticus). Four enzymatic hydrolysis systems were tested: an endopeptidase (Alcalase, A), an exopeptidase (Protana, P), two-stage hydrolysis with an endopeptidase followed by an exopeptidase (A + P), and a single stage with endo- and exopeptidase (AP). The results show that combined enzymatic treatments, especially single-stage Alcalase and Protana (AP), achieved high protein yields (80%) and enhanced degrees of hydrolysis (34 to 49%), producing peptides with lower molecular weights. FPH exhibited significant antioxidant activity, in 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) assays, with EC50 values below 5 mg/mL. Additionally, AP hydrolysates demonstrated over 60% angiotensin-converting enzyme (ACE) inhibition at 5 mg/mL, indicating potential antihypertensive applications. Antidiabetic and anti-Alzheimer activities were present, but at relatively low levels. AP hydrolysates, especially from gilthead seabream, proved to be the most promising. This study highlights the value of fish co-products as sources of functional peptides, contributing to waste reduction, and their potential applications in food, agriculture, and nutraceuticals.

The advances on the development of novel materials capable to enhance the shelf life of food products may contribute to reduce the current worldwide food waste problem. Zinc oxide nanoparticles (ZnO NPs) are considered GRAS (Generally Recognized as Safe) by the Food and Drug Administration (FDA) and due to their good antimicrobial properties are suitable to be applied as active compounds in food packaging. ZnO NPs were synthesized to be tested in active bionanocomposites through an eco-friendlier route using apple peel wastes. This work aimed to develop bionanocomposites based on chitosan and incorporated with ZnO NPs to characterize its bioactivity via in vitro and in situ studies, using fresh poultry meat as the food matrix. Overall, bio-based biodegradable films presented good antimicrobial activity, being the intrinsic antimicrobial properties of chitosan enhanced by the ZnO NPs added on the system. When used as primary packaging of the meat, the samples protected with the films presented a decrease on the deterioration speed, which was represented by the preservation of the initial reddish color of the meat and reduction on the oxidation process and microbiological growth. The nanoparticles enhanced especially the antioxidant properties of the films and proved to be potential food preservatives agents to be used in active food packaging.

Utilizing water resources rationally has become critical due to the expected increase in water scarcity. Cacti are capable of surviving with minimal water requirements and in poor soils. Despite being highly drought-resistant, cacti still faces limitations in realizing its full potential under drought-stress conditions. To this end, we investigated the interactive effect of humic substances (Hs) and arbuscular mycorrhizal fungi (AMF) on cactus plants under drought stress. In the study, a cactus pot experiment had three irrigation levels (W1: no irrigation, W2: 15% of field capacity, and W3: 30% of field capacity) and two biostimulants (Hs soil amendment and AMF inoculation), applied alone or combined. The findings show that the W1 and W2 regimes affected cactus performance. However, Hs and/or AMF significantly improved growth. Our results revealed that drought increased the generation of reactive oxygen species. However, Hs and/or AMF application improved nutrient uptake and increased anthocyanin content and free amino acids. Furthermore, the soil’s organic matter, phosphorus, nitrogen, and potassium contents were improved by the application of these biostimulants. Altogether, using Hs alone or in combination with AMF can be an effective and sustainable approach to enhance the tolerance of cactus plants to drought conditions, while also improving the soil quality.

Strategies to Improve the Barrier and Mechanical Properties of Pectin Films for Food Packaging: Comparing Nanocomposites with Bilayers

Traditional food packaging systems help reduce food wastage, but they also produce environmental impacts when not properly disposed of. Bio-based polymers are a promising solution to overcome these impacts, but they have poor barrier and mechanical properties. This work evaluates two strategies to improve these properties in pectin films: the incorporation of cellulose nanocrystals (CNC) or sodium montmorillonite (MMT) nanoparticles, and an additional layer of chitosan (i.e., a bilayer film). The bionanocomposites and bilayer films were characterized in terms of optical, morphological, hygroscopic, mechanical and barrier properties. The inclusion of the nanofillers in the polymer reduced the water vapor permeability and the hydrophilicity of the films without compromising their visual properties (i.e., their transparency). However, the nanoparticles did not substantially improve the mechanical properties of the bionanocomposites. Regarding the bilayer films, FTIR and contact angle studies revealed no surface and/or chemical modifications, confirming only physical coating/lamination between the two polymers. These bilayer films exhibited a dense homogenous structure, with intermediate optical and hygroscopic properties. An additional layer of chitosan did not improve the mechanical, water vapor and oxygen barrier properties of the pectin films. However, this additional layer made the material more hydrophobic, which may play an important role in the application of pectin as a food packaging material.

Algae and seaweeds are used in cookery since the beginnings of human civilization, particularly in several Asian cultures. Phenolic compounds are secondary metabolites produced by aquatic and terrestrial plants for their natural defense against external stimuli, which possess powerful antimicrobial and antioxidant properties that can be very important for the food industry. The main objective of this study was to develop a whey protein concentrate active coating, incorporated with a Fucus vesiculosus extract in order to delay the lipid oxidation of chicken breasts. Ten hydroethanolic extracts from F. vesiculosus were obtained and their antioxidant capacity was evaluated through two antioxidant activity assays: the DPPH radical scavenging activity and -carotene leaching assay. The total content in phenolics compounds was also determined by Folin-Ciocalteu method. The chosen extract was the one obtained from the freeze-dried F. vesiculosus using 75% (v/v) ethanol as extraction solvent. The extract was successfully incorporated into a whey protein film and successfully strengthened the thickness, tensile strength, and elastic modulus. The active film also was able to inhibit the chicken breasts lipid oxidation for 25 days of storage.

Phytochemicals Recovery from Grape Pomace: Extraction Improvement and Chemometric Study

In the last 20 years, an increased interest has been shown in the application of different types and combinations of enzymes to obtain phenolic extracts from grape pomace in order to maximize its valorization. Within this framework, the present study aims at improving the recovery of phenolic compounds from Merlot and Garganega pomace and at contributing to the scientific background of enzyme-assisted extraction. Five commercial cellulolytic enzymes were tested in different conditions. Phenolic compound extraction yields were analyzed via a Design of Experiments (DoE) methodology and a second extraction step with acetone was sequentially added. According to DoE, 2% w/w enzyme/substrate ratio was more effective than 1%, allowing a higher total phenol recovery, while the effect of incubation time (2 or 4 h) variation was more enzyme-dependent. Extracts were characterized via spectrophotometric and HPLC-DAD analyses. The results proved that enzymatic and acetone Merlot and Garganega pomace extracts were complex mixtures of compounds. The use of different cellulolytic enzymes led to different extract compositions, as demonstrated using PCA models. The enzyme effects were observed both in water enzymatic and in the subsequent acetone extracts, probably due to their specific grape cell wall degradation and leading to the recovery of different molecule arrays.

Quality Preservation and Shelf-Life Extension of Prickly Pear (Opuntia ficus-indica L. Mill) Using Edible Coatings

Prickly pear consumption is increasing across the world due to its rich variety of nutrients and bioactive compounds. Yet, it is a seasonal and highly perishable fruit, and the application of edible coatings emerges as an alternative to extend its shelf life. In this work, the effects of alginate, starch, chitosan, and pectin as coatings on the physicochemical, bioactive, microbiological, and textural properties of two prickly pear varieties (orange and red), kept under refrigeration (5 ± 2 °C) were evaluated for 6 weeks. Coatings proved to be helpful in the maintenance of the fruits’ color and textural properties, especially when pectin was applied. Overall, starch and chitosan can be considered the most effective coatings in preserving the quality of prickly pears among the options studied. A lower weight loss (8–10%) in fruits was achieved when starch and chitosan were applied, while in control fruits (without coating), the loss was 18–23%. Starch and chitosan also contributed to preserving the bioactivity of red fruits and showed good results in the preservation of total phenolic content in the orange fruits. In addition, starch and chitosan coatings also presented the best performance for the reduction of microbial contamination (both yeasts and molds and total mesophilic aerobic microorganisms). These findings highlight the role of edible coatings in preserving prickly pears, for a longer period, meeting consumers’ demand for fresh fruit.

Exploring the Benefits of Nutritional and Chemical Characteristics of Touriga Nacional and Arinto Varieties (Vitis vinifera L.)

Environmental degradation leads to an unsustainable food system. In addition to this issue, the consumption of foods that improve people’s health and well-being is recommended. One of the alternatives is undoubtedly the use of by-products of winemaking, namely in the form of grape pomace flour (GPF). To verify the benefits of using the Touriga Nacional and Arinto (Vitis vinifera L.) flour varieties, analytical determinations were made to identify and quantify different components. In terms of nutritional characterization, the Touriga Nacional GPF showed results that indicate better nutritional quality than the Arinto GPF. The Touriga Nacional and Arinto samples had protein contents of 10.13% and 8.38%, polyunsaturated fatty acids of 6.66% and 5.18%, soluble dietary fiber of 14.3% and 1.7%, and insoluble dietary fiber of 55.1% and 46.4%, respectively. The anthocyanins, proanthocyanidins, and flavonols presented in samples were detected by HPLC-DAD/ESI-MS. Atomic absorption spectrometry revealed elevated concentrations of certain elements in Touriga Nacional compared to Arinto, with the former showing higher levels of aluminum (130 mg/kg) and iron (146 mg/kg) against the latter’s Al (120 mg/kg) and Fe (112 mg/kg) content. GPF could become a valuable ingredient due to its nutritional quality and high content of various polyphenols.

Catalytic Upgrading of Microalgae-Based Bio-Oils for Sustainable Jet Fuel Production

The transition to sustainable energy systems has intensified the search for renewable alternatives to reduce greenhouse gas emissions and reliance on fossil fuels. In this context, microalgae have emerged as a promising third-generation feedstock for biofuel production due to their rapid development, high lipid content, and ability to grow in wastewater without competing with freshwater resources. In this study, the hydrotreatment of biocrudes derived from C. vulgaris, T. obliquus, and a mixed microalgal culture cultivated in domestic wastewater is investigated. Catalytic upgrading was applied using sulphided CoMo/Al2O3 (sCoMo) and Pt/Al2O3 catalysts. The results demonstrated that catalytic upgrading enhanced the upgraded bio-oils’ quality compared to non-catalysed reactions, confirming the crucial role of catalysts in improving bio-oil properties. Compared with the Pt catalyst, sCoMo produced higher yields of upgraded bio-oil, greater enrichment in carbon and hydrogen, and higher heating value (HHV), while effectively enhancing nitrogen and oxygen removal. However, when compared with the non-sulphided CoMo, the sulphiding treatment did not significantly improve denitrogenation and treated oil yields. The highest fraction of components within the jet fuel boiling range (37.7%) was obtained using a Pt catalyst, while the non-catalysed process yielded the lowest (26.6%). In this sense, catalytic upgrading of microalgae-based biocrude represents an important step towards the production of advanced and environmentally sustainable fuels.
