Salivary microbiome changes distinguish response to chemoradiotherapy in patients with oral cancer
Background Oral squamous cell carcinoma (SCC) is associated with oral microbial dysbiosis. In this unique study, we compared pre- to post-treatment salivary microbiome in patients with SCC by 16S rRNA gene sequencing and examined how microbiome changes correlated with the expression of an anti-microbial protein. Results Treatment of SCC was associated with a reduction in overall bacterial richness and diversity. There were significant changes in the microbial community structure, including a decrease in the abundance of Porphyromonaceae and Prevotellaceae and an increase in Lactobacillaceae . There were also significant changes in the microbial community structure before and after treatment with chemoradiotherapy, but not with surgery alone. In patients treated with chemoradiotherapy alone, several bacterial populations were differentially abundant between responders and non-responders before and after therapy. Microbiome changes were associated with a change in the expression of DMBT1, an anti-microbial protein in human saliva. Additionally, we found that salivary DMBT1, which increases after treatment, could serve as a post-treatment salivary biomarker that links to microbial changes. Specifically, post-treatment increases in human salivary DMBT1 correlated with increased abundance of Gemella spp., Pasteurellaceae spp . , Lactobacillus spp., and Oribacterium spp . This is the first longitudinal study to investigate treatment-associated changes (chemoradiotherapy and surgery) in the oral microbiome in patients with SCC along with changes in expression of an anti-microbial protein in saliva. Conclusions The composition of the oral microbiota may predict treatment responses; salivary DMBT1 may have a role in modulating the oral microbiome in patients with SCC. Tbx5 Buffers Inherent Left/Right Asymmetry Ensuring Symmetric Forelimb Formation
The forelimbs and hindlimbs of vertebrates are bilaterally symmetric. The mechanisms that ensure symmetric limb formation are unknown but they can be disrupted in disease. In Holt-Oram Syndrome (HOS), caused by mutations in TBX5, affected individuals have left-biased upper/forelimb defects. We demonstrate a role for the transcription factor Tbx5 in ensuring the symmetric formation of the left and right forelimb. In our mouse model, bilateral hypomorphic levels of Tbx5 produces asymmetric forelimb defects that are consistently more severe in the left limb than the right, phenocopying the left-biased limb defects seen in HOS patients. In Tbx hypomorphic mutants maintained on an INV mutant background, with situs inversus, the laterality of defects is reversed. Our data demonstrate an early, inherent asymmetry in the left and right limb-forming regions and that threshold levels of Tbx5 are required to overcome this asymmetry to ensure symmetric forelimb formation.

RADIATE – Radial Dysplasia Assessment, Treatment and Aetiology: protocol for the development of a core outcome set using a Delphi survey
Background Radial dysplasia (RD) is a disfiguring, potentially disabling congenital upper limb anomaly. Multiple surgical techniques are in current use, with little agreement on the optimal treatment approach. At present, no core outcome set exists specifically for RD, and the literature is dominated by retrospective case series. A recent systematic review by this group demonstrated significant heterogeneity on which outcomes are measured and how they are measured. Methods/design The RADIATE study will conduct a three-round online Delphi process, involving adult RD patients, the parents of children with RD, hand surgeons and hand therapists. The initial list of outcomes was drawn from our recent systematic review and will be supplemented by suggestions from the stakeholder groups. Following the Delphi process, outcomes that meet the consensus in definition will be ratified at a final consensus meeting. We will then follow the COSMIN guidelines to select outcome measurement instruments. Where appropriate, these will overlap with the outcome measures specified in the forthcoming standard set for congenital upper limb anomalies published by the International Consortium for Health Outcomes Measurement. Discussion The Radial Dysplasia Assessment, Treatment and Aetiology (RADIATE) study aims to address the uncertainty in the treatment of RD, and to begin to answer the question ‘What is the most appropriate treatment of the forearm and hand for children with RD?’ by establishing a core outcome set. Very regular high-frequency pulsation modes in young intermediate-mass stars
Asteroseismology probes the internal structures of stars by using their natural pulsation frequencies 1 . It relies on identifying sequences of pulsation modes that can be compared with theoretical models, which has been done successfully for many classes of pulsators, including low-mass solar-type stars 2 , red giants 3 , high-mass stars 4 and white dwarfs 5 . However, a large group of pulsating stars of intermediate mass—the so-called δ Scuti stars—have rich pulsation spectra for which systematic mode identification has not hitherto been possible 6 , 7 . This arises because only a seemingly random subset of possible modes are excited and because rapid rotation tends to spoil regular patterns 8 – 10 . Here we report the detection of remarkably regular sequences of high-frequency pulsation modes in 60 intermediate-mass main-sequence stars, which enables definitive mode identification. The space motions of some of these stars indicate that they are members of known associations of young stars, as confirmed by modelling of their pulsation spectra. The pulsation spectra of intermediate-mass stars (so-called δ Scuti stars) have been challenging to analyse, but new observations of 60 such stars reveal remarkably regular sequences of high-frequency pulsation modes.
TESS Observations of the Pleiades Cluster: A Nursery for δ Scuti Stars
We studied 89 A- and F-type members of the Pleiades open cluster, including five escaped members. We measured projected rotational velocities ( vsini ) for 49 stars and confirmed that stellar rotation causes a broadening of the main sequence in the color–magnitude diagram. Using time-series photometry from NASA’s TESS Mission (plus one star observed by Kepler/K2), we detected δ Scuti pulsations in 36 stars. The fraction of Pleiades stars in the middle of the instability strip that pulsate is unusually high (over 80%), and their range of effective temperatures agrees well with theoretical models. On the other hand, the characteristics of the pulsation spectra are varied and do not correlate with stellar temperature, calling into question the existence of a useful νmax relation for δ Scutis, at least for young main-sequence stars. By including δ Scuti stars observed in the Kepler field, we show that the instability strip is shifted to the red with increasing distance by interstellar reddening. Overall, this work demonstrates the power of combining observations with Gaia and TESS for studying pulsating stars in open clusters.
