Disseminated melanized fungal infection due to Cladosporium halotolerans in a dog coinfected with canine adenovirus-1 and canine parvovirus-2
This report presents the pathologic findings associated with disseminated infection due to Cladosporium halotolerans in a dog that was simultaneously infected with canine adenovirus-1 (CAdV-1) and canine parvovirus-2 (CPV-2). A 12-year-old, mixed breed dog, with a clinical history of neurological manifestations was submitted for routine autopsy due to poor prognosis. The principal pathologic findings were mycotic necrotizing nephritis, hepatitis, and splenitis with embolic dissemination to the brain resulting in mycotic necrotizing meningoencephalitis, ventriculitis, choroid plexitis, and obstructive hydrocephalus associated with intralesional and intravascular septate pigmented fungi. PCR and sequencing of the ITS region of fungi revealed that the intralesional fungal organisms had 82% nucleotide identity with members of the Cladosporium sphaerospermum complex of organisms. However, a PCR assay and sequencing of the beta tubulin gene confirmed that the organism identified in this dog had 100% nucleotide sequence identity with C. halotolerans . Using immunohistochemistry, intralesional antigens of CAdV-1 were identified within the epithelial cells of the liver and lungs; there was positive immunolabeling for CPV-2 antigens in degenerated cardiomyocytes. These findings confirmed the active participation of C. halotolerans in the development of disseminated cladosporiosis in this dog and represent a rare occurrence of concomitant infection with CAdV-1 and CPV-2.
Self-actualization and B-values: Development and validation of two instruments in the Brazilian context
Self-actualization is a complex psychological construct within Maslow’s motivation theory, characterized by numerous gaps in the empirical and measurement spectrums. Therefore, the objectives of this study are to develop, validate, and cross-verify measures for self-actualization attributes and B-values, focusing on job context and theoretical congruence with innovative behavior and human values related to the self-actualization construct (suprapersonal subfunction). This study involved a diverse sample of 621 Brazilian participants from 25 different professions, indicating the broad applicability of the findings. The proposed instruments underwent content and semantic validity assessments, followed by verification of factor validity and internal consistency. Results showed satisfactory content, semantic and factor validity and internal consistency parameters. The study reveals that self-actualization attributes can be understood through achieving one’s own potential and work meta-motivation, consistent with the adoption of B-values. Relationships with suprapersonal values (maturity, knowledge, and beauty) and innovative work behavior were also demonstrated, suggesting convergent validity evidence. The validation of SAAS and BVI contributes to understanding self-actualization and B-values in varied Brazilian contexts, offering insights for psychological assessment and intervention.
Estimating the impact of Industry 4.0 automation on curricular competence indicators in Brazilian vocational education and training: A mixed-methods AI-supported analysis
Context: The Fourth Industrial Revolution has accelerated the integration of automation technologies into the world of work, raising important questions about the future of Vocational Education and Training (VET). While existing literature has primarily focused on the labor market impacts of automation, few studies have investigated its direct effects on VET curricula. This article addresses this gap by assessing how automation may influence the structure and content of technical courses offered by Brazil's National Service for Commercial Apprenticeship (Senac), one of the country's largest VET providers. Approach: We implemented a three-stage methodology to estimate the impact of automation on technical education: (i) Technological mapping, (ii) prompt development, and (iii) assessment. In the third stage, we combined human expertise with generative Artificial Intelligence tools (GPT-4 and Claude 2) to evaluate 2,100 Course Competency Indicators (CCIs) across 35 technical courses. This dual approach enabled a scalable yet context-sensitive analysis, leveraging both the depth of human judgment and the efficiency of AI. Findings: The technological mapping identified seven key categories of automation technologies: 3D/4D Printing and Modeling, Applied AI, Data Analytics, Digital Platforms and Applications, Extended Reality, IoT and Connected Devices, and Robotics. The developed prompt provided structured guidance for assessing automation impact on CCIs, including instructions for classifying technologies, estimating impact levels, and justifying the results. The assessment showed that 70.3% of the CCIs are at Medium (39.1%) or Low (31.2%) levels of automation impact, suggesting that the courses remain current and relevant, challenging the narrative of rapid obsolescence in technical education. Digital Platforms and Applications were the most frequently cited technology, appearing nearly three times more often than Applied AI and Data Analytics. In contrast, 3D/4D Modeling and Extended Reality had limited relevance in the current course content. Conclusions: This research contributes to global discussions on the future of VET in the context of rapid technological change. It also highlights how automation risk assessments can support curriculum development by identifying where updates or innovations are most needed. Strengthening the alignment between training programs and emerging labor market demands will be essential to ensuring inclusive, future-oriented vocational education.
Analyzing the Challenges for Future Smart and Sustainable Cities
The fast growth of the urban population increases the demand for energy, water, and transportation, amongst other needs. This study explores the current state and future scenarios of Smart Cities and the environmental, economic, and social challenges that must be overcome. We used the Rapid Review method to understand the challenges of implementing Smart Cities in different urban contexts and the potential impact of research on Smart City planning in future Smart Cities. The study offers insights into the potential for Smart City growth while identifying obstacles that must be addressed to ensure sustainability. Results serve as a foundation for planning and decision-making, highlighting aspects such as the adoption of alternative energies, reduction in car use, preservation of ecosystems, waste reduction, citizen participation, infrastructure, and citizen data privacy, among others. These aspects are essential to overcome obstacles and promoting Smart Cities’ development.
A Serious Game (Health Unit in Focus) for Enhancing Undergraduate Education on Older Adults’ Health: Design and Validation Study
Population aging underlines the critical need to improve health professional training to adequately care for adults aged >60 years. Developing educational resources to support academics and professionals presents a valuable opportunity to enhance understanding of health conditions and improve clinical management. Serious games are designed to develop teaching, training, and learning skills. Their use in the educational setting is warranted, as they integrate digital aspects and gamification to create a playful experience for content acquisition. Deepening this theme in nursing education will improve assistance to the older adult population, leading to more qualified care based on gerontological practices and comprehensive health care for older adults. This study aims to develop and validate a serious game on older adult health for undergraduate nursing students. This quantitative and descriptive methodological study, conducted between February 2023 and December 2023 at a public university in the Federal District of Brazil, involved the active participation of 27 undergraduate nursing students in their eighth to tenth semesters. The game, Health Unit in Focus (HUF), was developed and validated with their input. It features 75 clinical cases distributed across 3 themes: pharmacology, metabolic syndrome, and semiology. Of the 40 students initially enrolled, 27 completed the study. The app was validated using the System Usability Scale and student feedback, and the results were reported following the Game-Based Intervention Reporting Guidelines (GAMING). The participants had a mean age of 22.67 (SD 1.44) years, were mostly female (20/27, 74%), and were in their eighth semester (26/27, 96%). The game received an average System Usability Scale score of 85.75 (median 86.57), classified as excellent, as all evaluated items scored >75. Participants considered the game easy to use; accessible; practical; and rich in well-founded, useful content. This high usability score, coupled with the overwhelmingly positive feedback from the students, instills confidence in the game's effectiveness. Furthermore, 100% (27/27) of students agreed that learning through games is effective and expressed interest in incorporating more interactive games into their training. The serious game HUF showed good usability, as its overall score was \"excellent,\" with its highest score in the odd-numbered items that addressed the positive aspects identified in the analysis. The serious game HUF is not just a valid and reliable tool for training nursing students but also an engaging and interactive approach to learning. Its ability to captivate and involve students in the learning process is a testament to its potential to revolutionize nursing education. It is essential that the development of new methodological resources, such as serious games, be based on scientific evidence to guarantee greater reliability and success in achieving their established objectives.
Previous BCG vaccination is associated with less severe clinical progression of COVID-19
Background BCG vaccination, originally used to prevent tuberculosis, is known to “train” the immune system to improve defence against viral respiratory infections. We investigated whether a previous BCG vaccination is associated with less severe clinical progression of COVID-19 Methods A case-control study comparing the proportion with a BCG vaccine scar (indicating previous vaccination) in cases and controls presenting with COVID-19 to health units in Brazil. Cases were subjects with severe COVID-19 (O2 saturation < 90%, severe respiratory effort, severe pneumonia, severe acute respiratory syndrome, sepsis, and septic shock). Controls had COVID-19 not meeting the definition of “severe” above. Unconditional regression was used to estimate vaccine protection against clinical progression to severe disease, with strict control for age, comorbidity, sex, educational level, race/colour, and municipality. Internal matching and conditional regression were used for sensitivity analysis. Results BCG was associated with high protection against COVID-19 clinical progression, over 87% (95% CI 74–93%) in subjects aged 60 or less and 35% (95% CI − 44–71%) in older subjects. Conclusions This protection may be relevant for public health in settings where COVID-19 vaccine coverage is still low and may have implications for research to identify vaccine candidates for COVID-19 that are broadly protective against mortality from future variants. Further research into the immunomodulatory effects of BCG may inform COVID-19 therapeutic research.
The Impact of the COVID-19 Pandemic on the Mental Health of Teachers and Its Possible Risk Factors: A Systematic Review
(1) Objective: The objective was to analyze the development of psychiatric pathologies/burnout syndrome and their possible risk factors in teachers in the context of the COVID-19 pandemic. (2) Methods: A qualitative systematic review was carried out, according to the PRISMA protocol, in the PubMed, Scopus, and Web of Science databases using a combination of the following descriptors [MeSH]: “mental health”, “mental disorders’’, “covid-19” and “school teachers’’. Articles selected were written in English, Portuguese and Spanish, published between November 2019 and December 2022. (3) Results: The most common psychiatric pathologies were generalized anxiety disorders and depression. Burnout syndrome was also quite prevalent. Of the 776 articles identified, 42 were selected after applying the eligibility criteria. Although there is variability among the analyzed studies, the risk factors most correlated with increased morbidity in teachers were: (i) being female; (ii) age below the fifth decade of life; (iii) pre-existence of chronic or psychiatric illnesses before the pandemic; (iv) difficulty in adapting to the distance education model; (v) family/work conflicts; (vi) negative symptoms caused by the pandemic. (4) Conclusions: Therefore, the COVID-19 impact on mental health appears to be more common in female teachers in their fifth decade of life and with pre-existing psychiatric comorbidities. However, prospective studies are needed to better map this situation.
Mucuna pruriens Administration Minimizes Neuroinflammation and Shows Anxiolytic, Antidepressant and Slimming Effects in Obese Rats
This study evaluated the effect of Mucuna pruriens (MP) administration on neuroinflammation and behavioral and murinometric parameters in obese rats. Proximate composition, oligosaccharide and phenolic compound profile of MP were determined. Wistar adult male rats were randomized into healthy (HG) and obese group (OG). The HG consumed a control chow diet while OG consumed a cafeteria diet for eight weeks. Then, they were subdivided into: Healthy (HG); Healthy with MP administration (HGMP); Obese (OG); Obese with MP administration (OGMP), with the consumption of the respective diets remaining for another eight weeks, in addition to gavage with MP extract to supplemented groups (750 mg/kg weight). MP presented a composition rich in proteins and phenolic compounds, especially catechin, in addition to 1-kestose and levodopa. Supplementation reduced food intake, body weight, and thoracic and abdominal circumferences in obese rats. MP showed anxiolytic and antidepressant effects and reduced morphological damage and expression of interleukin 6 in the hippocampus of obese rats. MP treatment showed satietogenic, slimming, anxiolytic and antidepressant effects, besides to minimizing hippocampal neuroinflammation in obese rats. Our results demonstrated the potential anti-obesity of MP which are probably related to the high content of bioactive compounds present in this plant extract.
Phytochemical Profile, Toxicity Evaluation and Antinociceptive Effect of the n-Butanolic Fraction from the Leaves of Calotropis procera (Aiton) W.T Aiton (Apocynaceae)
Calotropis procera, known as “Silk cotton”, stands out for the presence of various classes of bioactive compounds responsible for its ethnopharmacological properties. The study aimed to conduct a phytochemical investigation, evaluating the in vitro and in vivo toxicity together with the antinociceptive potential of an n-butanolic fraction (FB) from the leaves. The crude ethanolic extract (CEE) was obtained by maceration in ethanol for 72 h. It was then partitioned using a gradual solvent sequence. The FB was analyzed by HPLC-ESI-MS/MS in negative mode and 1H and 13C NMR. Toxicity was assessed by the erythrocyte hemolytic assay and acute oral toxicity test at a single dose of 300 mg·kg−1. The antinociceptive effect was assessed by the acetic acid-induced abdominal writhing test and the formalin test in mice at doses of 3.75, 7.5 and 15 mg·kg−1 per os. HPLC-ESI-MS/MS analysis identified flavonoids, phenolic acids, and the megastigmane roseoside, isolated for the first time in C. procera. The FB did not cause hemolytic effects or behavioral or physiological changes in mice. It showed an antinociceptive effect at all doses, reducing abdominal writhing by up to 91.46% and the licking time in phases 1 and 2 of the formalin test by up to 63.83% and 91.73%, respectively. In this study, it was possible to determine that an FB of a crude extract of C. procera leaves has antinociceptive activity, possibly associated with the phenolic compounds and roseoside found, with a lack of toxicity in vitro and in vivo, validating its ethnopharmacological use.
Staging liver fibrosis after severe yellow fever with ultrasound elastography in Brazil: A six-month follow-up study
Background Yellow fever (YF) is a hemorrhagic disease caused by an arbovirus endemic in South America, with recent outbreaks in the last years. Severe cases exhibit fulminant hepatitis, but there are no studies regarding its late-term effects on liver parenchyma. Thus, the aim of this study was to determine the frequency and grade of liver fibrosis in patients who recovered from severe YF and to point out potential predictors of this outcome. Methodology/Principal findings We followed-up 18 patients who survived severe YF during a recent outbreak (January-April 2018) in Brazil using ultrasound (US) with shear-wave elastography (SWE) at 6 months after symptoms onset. No patient had previous history of liver disease. Median liver stiffness (LS) was 5.3 (4.6-6.4) kPa. 2 (11.1%) patients were classified as Metavir F2, 1 (8.3%) as F3 and 1 (8.3%) as F4; these two last patients had features of cardiogenic liver congestion on Doppler analysis. Age and cardiac failure were associated with increased LS (p = 0.036 and p = 0.024, respectively). SAPS-3 at ICU admission showed a tendency of association with significant fibrosis ([greater than or equal to] F2; p = 0.053). 7 patients used sofosbuvir in a research protocol, of which none showed liver fibrosis (p = 0.119). Conclusions/Significance We found a low frequency of liver fibrosis in severe YF survivors. US with SWE may have a role in the follow up of patients of age and / or with comorbidities after hospital discharge in severe YF, a rare but reemergent disease.
