Inhibition of γ-glutamyl transpeptidase activity decreases intracellular cysteine levels in cervical carcinoma
To determine whether gamma-glutamyl transpeptidase (gamma-GT) is involved in the maintenance of elevated cysteine levels in cervical carcinoma. Four cervical carcinoma cell lines were tested in vitro for cysteine accumulation and gamma-GT levels. The highest and lowest gamma-GT-expressing cell lines were used in in vivo experiments to determine the effect of gamma-GT inhibition on cysteine levels. Treatment of a series of cervical carcinoma cell lines with acivicin decreased intracellular cysteine concentrations. Cysteine depletion was evident in Me180 cells which had the greatest levels of gamma-GT activity, and had a more pronounced cysteine decrease in medium with glutathione and cysteine concentrations simulating the in vivo situation. Also investigated were the effects of inhibition of gamma-GT activity on intracellular cysteine levels in xenografts grown in severe combined immunodeficient (SCID) mice. With the use of 35 mg/kg of acivicin, gamma-GT activity decreased to basal levels of detection in both tumour types and significant decreases in cysteine levels were seen in the high gamma-GT-expressing tumours (Me180). Thus, inhibition of gamma-GT activity may have therapeutic potential in high-expressing cancers. In tumours and cell lines with elevated levels of gamma-GT activity, inhibition of this enzyme led to decreases of cysteine levels.

Reading Ecosystem Services at the Local Scale through a Territorial Approach
In recent years, the ecosystem services (ES) concept has become a major paradigm for natural resource management. While policy-makers demand “hard” monetary evidence that nature conservation would be worth investing in, ongoing attempts are being made to formalize the concept as a scientifically robust “one size fits all” analytical framework. These attempts have highlighted several major limitations of the ES concept. First, to date, the concept has paid little attention to the role of humans in the production of ES. Second, the ongoing formalization of the ES concept is turning it into a “technology of globalization,” thereby increasingly ignoring the socio-cultural context and history within which ecosystems emerge. Third, economic valuation has been shown to limit local stakeholders in expressing their daily and immediate ways of interacting with their environment over and beyond extrinsic motivation provided by financial gains. We address these three limitations by analyzing a social evaluation of the roles of peri-urban farmland from a territorial perspective. Our case study is the Thau lagoon in southern France. We conducted in-depth interviews with a broad range of stakeholders and ran two participatory workshops. Using a territorial meta-model that distinguishes three levels—physical, logical, and existential—stakeholder data were analyzed to unravel the interplay of territorial elements at these three levels that gives rise to ES in two broad categories: food production and aesthetic landscape. The coupling of ES and territory concepts opens up several novel analytical perspectives. It allows partitioning of ES in a manner that “re-contextualizes” them and gives insight about both their physical constituents and their meaning at the territorial level. Additional research should incorporate the dynamics of service demand and supply, and further investigate options for implementation.
Axion dark matter detection by laser spectroscopy of ultracold molecular oxygen: a proposal
Generalizing Sikivie's approach (Sikivie 2014 Phys. Rev. Lett.113 201301), according to which dark matter axions may induce transitions between Zeeman states in an atomic system, an experiment based on a molecular gas is proposed for a search in the axion mass range between 1.4 and 1.9 meV. In particular, a 16O2 sample is brought to the temperature of 280 mK via buffer-gas cooling and then subjected to an external magnetic field; axion-driven transitions are eventually detected by resonance-enhanced multi-photon ionization spectroscopy.
Molecular Profiling of Spermatozoa Reveals Correlations between Morphology and Gene Expression: A Novel Biomarker Panel for Male Infertility
Choosing spermatozoa with an optimum fertilizing potential is one of the major challenges in assisted reproductive technologies (ART). This selection is mainly based on semen parameters, but the addition of molecular approaches could allow a more functional evaluation. To this aim, we used sixteen fresh sperm samples from patients undergoing ART for male infertility and classified them in the high-and poor-quality groups, on the basis of their morphology at high magnification. Then, using a DNA sequencing method, we analyzed the spermatozoa methylome to identify genes that were differentially methylated. By Gene Ontology and protein-protein interaction network analyses, we defined candidate genes mainly implicated in cell motility, calcium reabsorption, and signaling pathways as well as transmembrane transport. RT-qPCR of high-and poorquality sperm samples allowed showing that the expression of some genes, such as AURKA, HDAC4, CFAP46, SPATA18, CACNA1C, CACNA1H, CARHSP1, CCDC60, DNAH2, and CDC88B, have different expression levels according to sperm morphology. In conclusion, the present study shows a strong correlation between morphology and gene expression in the spermatozoa and provides a biomarker panel for sperm analysis during ART and a new tool to explore male infertility.
Energy Recovery Based on Exhaust Gas Recirculation and Heat Regeneration Processes Applied in a Firewood Boiler
This research investigated the exhaust gas recirculation (EGR) process to recover part of the thermal and chemical energy left in the exhaust boiler stream. A theoretical energy conversion and use analysis was performed based on a small boiler. Several measurements and analyses of the operation reports provided the boundary conditions and relevant information for modelling the processes. The methodology considered the radiation from exhaust gases, thermodynamics balances, and financial engineering calculations for the energy recovery analysis. Financial results indicate that the exhaust gas recirculation process implementation, regarding 20% of the EGR ratio, presented 69% and 1.45 years of internal returning rate and payback, respectively. However, the regenerative process presented an internal returning rate and payback values of 112% and 0.9 years. Indeed, both processes might be applied in order to increase efficiency and reduce emissions.

An experimental apparatus for measuring the Casimir effect at large distances
An experimental set-up for the measurement of the Casimir effect at separations larger than a few microns is presented. The apparatus is based on a mechanical resonator and uses a homodyne detection technique to sense the Casimir force in the plane-parallel configuration. First measurements in the 3-10 micrometer range show an unexpected large force probably due to patch effects.
Search of spin-dependent fifth forces with precision magnetometry
Spin-dependent fifth-forces are associated with particles beyond the standard model. In particular, light pseudo-scalar bosons mediate long-range forces, allowing mass to interact with spins. The search of these interactions can be performed by periodically varying the distance between a source mass and a spin ensemble, in order to modulate the force intensity and detect it with precision magnetometry techniques. In our setup the force arises from room temperature lead masses and is detected in a paramagnetic crystal at 4.2\\,K, whose magnetisation is monitored by a SQUID-based magnetometer with the sensitivity of \\(53\\,aT/Hz\\). Our measurement places the most stringent constraints on a spin-mass interaction in the ranges 1\\,cm to 10\\,m and 10\\,km to 300\\,km, improving existing limits up to more than two orders of magnitude. We show that this experimental technique may be further leveraged to explore a vast region of the fifth force's parameter space, with an interaction range longer than a few centimetres.
