Themed Project Case Study: Quadruple Tanks Control with PLCs
A themed project based on the control of a quadruple tank rig using PLCs has been successfully carried out as part of the MSc in Advanced Control and Systems Engineering at the University of Manchester. The themed project involves ten students who address a single multivariable control challenge under the supervision of two academics and four PhD students. As every student is required to write their own MSc dissertation, the key point is the possibility of using different control techniques to be implemented on different hardware platforms.
Use of yeast (Pichia kudriavzevii) as a novel feed additive to ameliorate the effects of aflatoxin B1 on broiler chicken performance
The aim of this study was to evaluate the efficacy of autochthonous Pichia kudriavzevii as a novel bioadsorbent for aflatoxin B 1 (AFB 1 ). The selection of this yeast was based on the AFB 1 adsorption capacity previously demonstrated in vitro (Magnoli et al. 2016 ). One-day-old Cobb broilers ( n  = 160) were randomly assigned to four dietary treatments (T1: basal diet (B); T2: B + 0.1% yeast; T3: B + AFB 1 , 100 μg/kg; T4: B + 0.1% yeast + AFB 1 , 100 μg/kg). Performance parameters (average daily weight gain body, average daily consumption, feed conversion ratio, carcass weight, and dead weight), biochemical parameters (albumin, globulin, and albumin/globulin), liver pathological changes, and AFB 1 residual levels in the liver and excreta were evaluated. Significant differences ( P  < 0.05) in performance parameters were observed among treatments and controls: T3 group showed the lowest average daily body weight gain value while in T4 group, the value of this parameter increased significantly ( P  < 0.05). T3 and T4 groups showed the lowest and highest values for average daily feed consumption, respectively. The feed conversion ratio (FC) showed no significant differences among treatments. T3 group showed the lowest dead weight and carcass weight compared with T1 group. The biochemical parameters showed no significant differences among treatments. T3 group showed macroscopic and microscopic liver changes compared to the control. Aflatoxin B 1 levels (μg/g) were detected in broiler livers and showed significant differences among treatments ( P  < 0.05). In conclusion, native P. kudriavzevii incorporation (0.1%) in broiler diets containing AFB 1 was shown to be effective in ameliorating the adverse effects of AFB 1 on production.
Porcine colonization of the Americas: a 60k SNP story
The pig, Sus scrofa, is a foreign species to the American continent. Although pigs originally introduced in the Americas should be related to those from the Iberian Peninsula and Canary islands, the phylogeny of current creole pigs that now populate the continent is likely to be very complex. Because of the extreme climates that America harbors, these populations also provide a unique example of a fast evolutionary phenomenon of adaptation. Here, we provide a genome wide study of these issues by genotyping, with a 60k SNP chip, 206 village pigs sampled across 14 countries and 183 pigs from outgroup breeds that are potential founders of the American populations, including wild boar, Iberian, international and Chinese breeds. Results show that American village pigs are primarily of European ancestry, although the observed genetic landscape is that of a complex conglomerate. There was no correlation between genetic and geographical distances, neither continent wide nor when analyzing specific areas. Most populations showed a clear admixed structure where the Iberian pig was not necessarily the main component, illustrating how international breeds, but also Chinese pigs, have contributed to extant genetic composition of American village pigs. We also observe that many genes related to the cardiovascular system show an increased differentiation between altiplano and genetically related pigs living near sea level.
Experimental Evaluation of the Effectiveness Offered by Different Types of Personal Protective Clothing Against Nanoaerosols
The rapid expansion of nanotechnology is outpacing health and safety recommendations for engineered nanomaterials. Thus, there is a lack of information about the effects that nanomaterials can induce in the human health. Nevertheless, workers in nanotechnology-related industries are potentially at risk of being exposed to nanomaterials. Therefore, there is a need of characterize the behaviour of personal protective equipment against penetration nanoparticles, in order to provide an adequate protection to the workers. In this study, the efficiency of several protective dermal equipment against water-based NaCl aerosol was evaluated. For this purpose, different protective clothing and gloves were selected to carry out the assays, simulating typical use conditions of protective equipment under occupational settings. Results obtained exposed that the level of protection offered by the distinct types of personal protective coveralls depended not only on the fabric, but also on their fitting to the body of the subject. On the other hand, the efficiency of the protective gloves was set in the range from 95% to 99%, depending on the thickness and the type of material.
Epistatic effects occurring among susceptibility and protective MHC genes in IgA deficiency
Immunoglobulin A deficiency (IgAD), the most prevalent primary immunodeficiency in Caucasian populations, shows strong evidence of polygenic inheritance with several associated genes being located in the major histocompatibility complex (MHC). Our aims were to determine which previously described MHC associations were primary and not secondary to a decrease or an increase in other MHC haplotype frequencies, to study the genetic interactions between all disease-associated MHC haplotypes and, finally, to ascertain the relative importance of protection vs susceptibility. A relative predispositional effect (RPE) study showed that in addition to the primary positive association of IgAD with HLA-DRB1 * 0102, DR3/TNFa2b3, and DR7 carrying haplotypes, DRB1 * 1501 was a marker of a primary protective factor in the Spanish population. Our data also indicate that the combined presence in an individual of two MHC susceptibility haplotypes notably increases the predisposition to the disease and that DRB1 * 1501 positive haplotypes eliminate the susceptibility conferred by any other MHC haplotype.
Effect of Recombinant Prophenin 2 on the Integrity and Viability of Trichomonas vaginalis
Trichomonas vaginalis is the causal agent of trichomoniasis, which is associated with preterm child delivery, low birth weight, and an increased risk of infection by human papilloma virus and human immunodeficiency virus following exposure. Several reports have established increasing numbers of trichomoniasis cases resistant to metronidazole, the agent used for treatment, and it is therefore important to identify new therapeutic alternatives. Previously, our group reported the effect of tritrpticin, a synthetic peptide derived from porcine prophenin, on T. vaginalis; however, the hemolytic activity of this small peptide complicates its possible use as a therapeutic agent. In this study, we report that the propeptide and the processed peptide of prophenin 2 (cleaved with hydroxylamine) affected the integrity and growth of T. vaginalis and that pro-prophenin 2 displays some resistance to proteolysis by T. vaginalis proteinases at 1 h. Its effect on T. vaginalis as well as its low hemolytic activity and short-time stability to parasite proteinases makes prophenin 2 an interesting candidate for synergistic or alternative treatment against T. vaginalis.
