Induction of resident memory T cells enhances the efficacy of cancer vaccine
2017
Tissue-resident memory T cells (Trm) represent a new subset of long-lived memory T cells that remain in tissue and do not recirculate. Although they are considered as early immune effectors in infectious diseases, their role in cancer immunosurveillance remains unknown. In a preclinical model of head and neck cancer, we show that intranasal vaccination with a mucosal vector, the B subunit of Shiga toxin, induces local Trm and inhibits tumour growth. As Trm do not recirculate, we demonstrate their crucial role in the efficacy of cancer vaccine with parabiosis experiments. Blockade of TFGβ decreases the induction of Trm after mucosal vaccine immunization, resulting in the lower efficacy of cancer vaccine. In order to extrapolate this role of Trm in humans, we show that the number of Trm correlates with a better overall survival in lung cancer in multivariate analysis. The induction of Trm may represent a new surrogate biomarker for the efficacy of cancer vaccine. This study also argues for the development of vaccine strategies designed to elicit them.
Resident memory T cells (Trm) are memory T cells that remain in tissue. Here, the authors show that induction of Trm cells is required for control of tumour growth following mucosal vaccination in mice bearing head and neck cancer and that Trm cells in human lung cancer correlates with a better survival.

American Sign Language Alphabet Recognition Using a Neuromorphic Sensor and an Artificial Neural Network

This paper reports the design and analysis of an American Sign Language (ASL) alphabet translation system implemented in hardware using a Field-Programmable Gate Array. The system process consists of three stages, the first being the communication with the neuromorphic camera (also called Dynamic Vision Sensor, DVS) sensor using the Universal Serial Bus protocol. The feature extraction of the events generated by the DVS is the second part of the process, consisting of a presentation of the digital image processing algorithms developed in software, which aim to reduce redundant information and prepare the data for the third stage. The last stage of the system process is the classification of the ASL alphabet, achieved with a single artificial neural network implemented in digital hardware for higher speed. The overall result is the development of a classification system using the ASL signs contour, fully implemented in a reconfigurable device. The experimental results consist of a comparative analysis of the recognition rate among the alphabet signs using the neuromorphic camera in order to prove the proper operation of the digital image processing algorithms. In the experiments performed with 720 samples of 24 signs, a recognition accuracy of 79.58% was obtained.

This work illustrates the analysis of Film Bulk Acoustic Resonators (FBAR) using 3D Finite Element (FEM) simulations with the software OnScale in order to predict and improve resonator performance and quality before manufacturing. This kind of analysis minimizes manufacturing cycles by reducing design time with 3D simulations running on High-Performance Computing (HPC) cloud services. It also enables the identification of manufacturing effects on device performance. The simulation results are compared and validated with a manufactured FBAR device, previously reported, to further highlight the usefulness and advantages of the 3D simulations-based design process. In the 3D simulation results, some analysis challenges, like boundary condition definitions, mesh tuning, loss source tracing, and device quality estimations, were studied. Hence, it is possible to highlight that modern FEM solvers, like OnScale enable unprecedented FBAR analysis and design optimization.

Chlorhexidine-Induced Contact Anaphylaxis

Perioperative anaphylaxis is a rare but life-threatening event, with chlorhexidine increasingly recognized as a significant trigger. Though commonly used as an antiseptic due to its broad-spectrum antimicrobial properties, chlorhexidine can lead to sensitization through repeated exposure, resulting in both immediate and delayed hypersensitivity reactions. We report a case of severe perioperative anaphylaxis in a 72-year-old woman following abdominal surgery at the University Hospital of Besançon in May 2024. Chlorhexidine was used for preoperative skin antisepsis. Shortly after anesthesia induction with etomidate and rocuronium, the patient developed bronchospasm, followed by refractory hypotension and oxygen desaturation after administration of additional anesthetics. She required intensive care admission. Elevated acute serum tryptase (123 µg/L) confirmed anaphylaxis. Allergy investigations revealed positive skin tests and specific immunoglobulin E (IgE) to chlorhexidine, while tests for other agents were negative. Genetic testing excluded mast cell disorders or hereditary alpha-tryptasemia. Surgery was successfully completed five days later with chlorhexidine avoidance and no further reactions. This case underscores the potential severity of chlorhexidine-induced perioperative anaphylaxis. Given the widespread use of chlorhexidine and the possibility of delayed or underrecognized sensitization, clinicians must remain vigilant, particularly in patients with prior unexplained reactions. Preoperative screening and heightened awareness are essential to reduce morbidity and mortality associated with this increasingly reported allergen.

América Latina: gobernanza y el recurso vital

At present the world socioeconomic system is at a great crossroads, the New World Order is in the decisive offensive to achieve its objectives and it is not known if it will succeed. The military industrial complex, negative accumulation of capital based on the destructive sectors, and the productive only as their sustenance, which implemented the empire, is no longer the key to the extermination of the population, which is the product of a change in strategy generated by the end of the cold war. Here the issue of governance based on current indicators of a Latin American sub-region: Ecuador, Bolivia and Venezuela and the challenges that the other subregion faces Brazil, Argentina, Mexico, Chile, Peru and Paraguay, while the Caribbean is going behind. And in the last part we can see the new vital juncture, water stress in Latin America and its inequalities, despite being a continent with sufficient but predatory water resources. Issues that affect mortality but do not compare with Asia and Africa, culminating with a reflection.

La violencia en Guerrero: factores históricos, estructurales, coyunturales e inerciales
Desde la perspectiva anterior, se ha considerado en este estudio la situation no solo marginal que ha vivido historicamente Guerrero, sino tambien dentro de los avances de las ciencias sociales, las relaciones encontradas entre la desigualdad social y el incremento de la violencia en America Latina, y particularmente en Mexico, donde subsiste la falta de oportunidades para acceder a empleos bien remunerados cuyos soportes sean los meritos para el progreso socioeconomico de las personas, y que el medio para lograr esto sea el impulso a la education mas no el enriquecimiento y concentration de la riqueza de unos cuantos por la via de herencias incrementadas por favorecimientos de las estructuras poh'ticas, como ha sucedido historicamente en Mexico, pero con mayor acentuacion desde la puesta en marcha del neoliberalismo. Sin embargo, habra que ir mas alia de los intereses politicos y articular estos a un interes mayor desde la perspectiva del dominio economico regional, pues en las principales ciudades guerrerenses las familias de estos grandes caciques mantienen grandes patrimonies y negocios que pretenden acrecentar aun mas, al amparo del protagonismo politico en el cual ya desde hace varias decadas han estado inmersos. A Io anterior, habra que agregar que en Guerrero se ha venido configurando una estrategia de incursionar en la poh'tica como medio para prosperarsocioeconomicamente y subir de estatus social; incluso esto ha ido permeando en el imaginario colectivo de las nuevas generaciones de jovenes cuyos padres o abuelos pertenecen a alguna estirpe poh'tica de hace mucho tiempo, aunque tambien se ha hecho cada vez mas notorio el rechazo a quienes hacen de la poh'tica una profesion de enriquecimiento para trepar por encima del pueblo, Io cual ha desprestigiado a quienes participan para cargos publicos de election popular, y es asi que actualmente en el numero de asesinatos de politicos por entidadfederativa, Guerrero figure en primer lugar con 10 hasta el 8 de marzo de 2018 (Proceso, con sustento en datos de Etellekt Consuftores, 2018), y se han incrementado exponencialmente los asesinatos de personas vinculadas con la poh'tica tanto estatal como nacional. Asi', ante una estructura social que impide el camino para jovenes que desean superar su condition social, se explica que muchos de ellos busquen espacios en la poh'tica a riesgo de perder sus vidas, en un contexto socioeconomico y cultural donde al amparo de la ideologi'a de la globalization se busca justificar el rechazo a muchos jovenes que han buscado la preparation profesional, pero que bajo el argumento de que no son aptos como capital humano para el empleo y la remuneration a que aspiran, son excluidos.

A Reduced Hardware SNG for Stochastic Computing

Stochastic Computing (SC) is an alternative way of computing with binary weighted words that can significantly reduce hardware resources. This technique relies on transforming information from a conventional binary system to the probability domain in order to perform mathematical operations based on probability theory, where smaller amounts of binary logic elements are required. Despite the advantage of computing with reduced circuitry, SC has a well known issue; the input interface known as stochastic number generator (SNG), is a hardware consuming module, which is disadvantageous for small digital circuits or circuits with several input data. Hence, in this work, efforts are dedicated to improving a classic weighted binary SNG (WBSNG). For this, one of the internal modules (weight generator) of the SNG was redesigned by detecting a pattern in the involved signals that helped to pose the problem in a different way, yielding equivalent results. This greatly reduced the number of logical elements used in its implementation. This pattern is interpreted with Boolean equations and transferred to a digital circuit that achieves the same behavior of a WBSNG but with less resources.
