Monitoring the efficacy of antimalarial medicines in India via sentinel sites: Outcomes and risk factors for treatment failure
Background & objectives: To combat the problem of antimalarial drug resistance, monitoring the changes in drug efficacy over time through periodic surveillance is essential. Since 2009, systematic and continuous monitoring is being done through nationwide sentinel site system. Potential early warning signs like partner drug resistance markers were also monitored in the clinical samples from the study areas. Methods: A total of 1864 patients with acute uncomplicated malaria were enrolled in therapeutic efficacy studies of artesunate plus sulphadoxine-pyrimethamine (AS+SP) for Plasmodium falciparum; those infected with P. vivax were given chloroquine (CQ). Polymerase chain reaction (PCR) was used to distinguish post-treatment reinfection from treatment failures. Isolates of P. falciparum were also analysed for dihydropteroate synthase (dhps) and dihydrofolate reductase (dhfr) gene mutations. Results: Overall, 1687 (91.7%) patients completed the follow-up. In most of the falciparum patients the parasitaemia was cleared within 24 h of treatment, except 12 patients who remained parasite positive after 72 h. Presence of dhfr and dhps quintuple mutation was observed predominantly in treatment failure samples. A daily dose of artesunate of < 3 mg/kg of body weight, age of <5 yr, and fever at enrolment were associated with an increased risk of treatment failure. The AS+SP in P. falciparum was effective in > 95% cases in all the sentinel sites except in Northeastern region (NE). Chloroquine remained 100% efficacious in case of P. vivax infections. Interpretation & conclusion: Till 2012, India′s national antimalarial drug resistance monitoring system proved highly efficacious and safe towards first-line antimalarials used in the country, except in Northeastern region where a decline in efficacy of AS+SP has been observed. This led to change in first-line treatment for P. falciparum to artemether-lumefantrine in Northeastern region.
The Role of Instability Indices in Forecasting Thunderstorm and Non-Thunderstorm Days across Six Cities in India
Thunderstorms are one of the most damaging natural hazards demanding in-depth understanding and prediction. These convective systems form in an unstable environment which is quantitatively expressed in terms of instability indices. These indices are studied over six locations across the Indian landmass in an attempt to predict thunderstorm activity on any given day. A combination of multiple regression, logistic regression, and range analysis provides new insight into the prediction of these storms. A supervised machine learning-based logistic regression model is developed in this study for thunderstorm prediction over Patna and can be further extended for operational forecasting of Thunderstorms over the region. Critical thresholds for the instability indices are determined over the considered locations providing valuable insight into the domain of Thunderstorm prediction
Impact of Export and Import Dynamics on the Balance of Trade for Turmeric-Producing Countries: A Markov Chain Analysis
The study reveals significant fluctuations in turmeric area, production, yield, and trade patterns over time. From 1950 to 2023, average area and output increased by 2.72% and 4.05%, respectively, while yield growth remained at 1.32%. The average production value peaked in Period 4 at 1031.84 before falling to 129.11 in Period 5. The coefficient of variation increased sharply from 16.62 to 81.44 in Period 5, indicating greater unpredictability and variability. From 2015 to 2024, Indias turmeric trade experienced fluctuations in export growth, increasing by 39.6% in 2016 and 54.1% in 2018, but decreasing by 17.8% in 2017 and 46.1% in 2024. Germany, Ethiopia, and Myanmar are important hubs in Indias turmeric export network. Effective strategies are needed to support sustainable practices, increase value chain efficiency, and improve market access. Policy focus for turmeric cultivation in 2024 should focus on supporting organic farming, investing in research to develop disease-resistant, high-yield varieties, strengthening local processing facilities, implementing cluster-based sustainable development, and integrating digital platforms for direct market access and improved export logistics.
Emerging polymorphisms in falciparum Kelch 13 gene in Northeastern region of India
Background Recent reports of emergence and spread of artemisinin resistance in the Southeast Asia region, including Myanmar, pose a greater threat to malaria control and elimination in India. Whole genome sequencing studies have associated mutations in the K13 propeller gene ( k13 ), PF3D7_1343700 with artemisinin resistance both in vitro and in vivo. The aim of the present study was to find the k13 gene polymorphisms in Plasmodium falciparum parasites from the three sites in the Northeast region of India, bordering Bangladesh and Myanmar. Methods A total of 254 samples collected during 2014–2015 from Tripura, Mizoram and Arunachal Pradesh states in the Northeast region of India were used to obtain the full-length k13 gene sequences. Results Three non-synonymous (NS) mutations: two in the propeller region, namely at codon 446 and 578, were observed besides one at codon 189 in the non-propeller region. The treatment outcome was not affected by these mutations at any of the sites. In addition, microsatellite variation in the N-terminus of the k13 protein was observed at all the study sites. Conclusion This is the first study to document the presence of F446I NS mutation in the k13 propeller region from Changlang district, Arunachal Pradesh, a site adjoining the Indo-Myanmar border region, where this mutation is highly prevalent. In addition, NS mutation A578S has been observed only at Lunglei district, Mizoram, a site bordering Bangladesh and K189T mutation with relatively higher frequency in Mizoram and Tripura states. The presence of F446I mutation in a region close to the Myanmar border is notable. Considering the spread of anti-malarial drug resistance from Southeast Asia to the Northeast region of India in the past, there is an urgent need to undertake systematic mapping studies to ascertain the role and extent of this mutation in artemisinin resistance in this region of country.
Declining efficacy of artesunate plus sulphadoxine-pyrimethamine in northeastern India
Background Anti-malarial drug resistance in Plasmodium falciparum in India has historically travelled from northeast India along the Myanmar border. The treatment policy for P. falciparum in the region was, therefore, changed from chloroquine to artesunate (AS) plus sulphadoxine-pyrimethamine (SP) in selected areas in 2005 and in 2008 it became the first-line treatment. Recognizing that resistance to the partner drug can limit the useful life of this combination therapy, routine in vivo and molecular monitoring of anti-malarial drug efficacy through sentinel sites was initiated in 2009. Methods Between May and October 2012, 190 subjects with acute uncomplicated falciparum malaria were enrolled in therapeutic efficacy studies in the states of Arunachal Pradesh, Tripura, and Mizoram. Clinical and parasitological assessments were conducted over 42 days of follow-up. Multivariate analysis was used to determine risk factors associated with treatment failure. Genotyping was done to distinguish re-infection from recrudescence as well as to determine the prevalence of molecular markers of antifolate resistance among isolates. Results A total of 169 patients completed 42 days of follow-up at three sites. The crude and PCR-corrected Kaplan-Meier survival estimates of AS + SP were 60.8% (95% CI: 48.0-71.4) and 76.6% (95% CI: 64.1-85.2) in Gomati, Tripura; 74.6% (95% CI: 62.0-83.6) and 81.7% (95% CI: 69.4-89.5) in Lunglei, Mizoram; and, 59.5% (95% CI: 42.0-73.2) and 82.3% (95% CI: 64.6-91.6) in Changlang, Arunachal Pradesh. Most patients with P. falciparum cleared parasitaemia within 24 hours of treatment, but eight, including three patients who failed treatment, remained parasitaemic on day 3. Risk factors associated with treatment failure included age < five years, fever at the time of enrolment and AS under dosing. No adverse events were reported. Presence of dhfr plus dhps quintuple mutation was observed predominantly in treatment failure samples. Conclusion AS + SP treatment failure was widespread in northeast India and exceeded the threshold for changing drug policy. Based on these results, in January 2013 the expert committee of the National Vector Borne Disease Control Programme formulated the first subnational drug policy for India and selected artemether plus lumefantrine as the new first-line treatment in the northeast. Continued monitoring of anti-malarial drug efficacy is essential for effective malaria control.
Economics of Lemongrass Cultivation in Jharkhand State
The present study was carried out to study the cost of cultivation and returns from lemongrass, oil bearing aromatic crop using different cost concepts, estimated from primary data collected from 75 cultivators. The cost of cultivation was calculated using operating expenses (Cost A1) at ` 51,713, while considering all types of costs (Cost C3) it reached to ` 68691 per hectare. The gross returns from cultivation of lemongrass in a hectare was found at ` 1,40,000 hectare, while net return over Cost A1 and Cost C3 was estimated at ` 71309 and ` 88287, respectively. The B: C ratio over cost A1 and cost C3 was calculated as ` 1.71 and ` 1.04, which indicated that lemongrass cultivation is profitable venture in the study area. Estimation of a regression model indicated positive and significant influence of expenditure on machine use, lemongrass slips and manures/ fertilizer on returns from lemongrass. There are two channels for marketing of lemongrass oil, i.e. (i) producer-local buyer-industry, and (ii) producer- industry. It was observed that marketing cost and price spread was higher in channel-I due to more number of intermediaries. The lemongrass cultivator can ease their marketing activities and enhance income through formation of cooperative society.
Exploring the benefits of biochar over other organic amendments for reducing of metal toxicity in Withania somnifera
AbstractBiochar is considered as a good metal sequester and ameliorates the metal toxicity and uptake in plants. However, its effectiveness over other organic amendments is not well discussed. The aim of this study is to compare the biochar amendments alleviating the levels of Pb and Cd in Withania somnifera L. Dunal with other organic manure. Farmyard manure, vermicompost, and biochar were applied in the field spiked with highly available Pb and Cd. Metal bioavailability and accumulation, plant growth and plant physiology, antioxidant enzymes and metabolite content of W. somnifera along with soil properties were evaluated in all treatments. Study indicates that the application of organic manures significantly alleviated the metal levels (33–72%) in the plant in comparison with control. The herbage yield was significantly higher (33–69%) under different organic manures as compared to the metal treatments. Secondary metabolite contents and antioxidant enzymes were higher in metal treatments in comparison with control. Organic manures were not only able to restrain the Pb and Cd in soil but also enhanced the soil microbial activities. Results indicate that biochar amendments were more promising than farmyard manure and vermicompost due to the presence of more stable carbon in biochar and more alleviation in metal uptake. In addition, net profit in the cultivation of W. somnifera was higher for biochar amendments (50% higher) compared to control. The study recommended that biochar could be a better option for commercial and safer production of W. somnifera.Graphic abstract
Towards the development of phytoextract based healthy ageing cognitive booster formulation, explored through Caenorhabditis elegans model
The positive effect of herbal supplements on aging and age-related disorders has led to the evolution of natural curatives for remedial neurodegenerative diseases in humans. The advancement in aging is exceedingly linked to oxidative stress. Enhanced oxidative stress interrupts health of humans in various ways, necessitating to find stress alleviating herbal resources. Currently, minimal scientifically validated health and cognitive booster resources are available. Therefore, we explored the impact of plant extracts in different combinations on oxidative stress, life span and cognition using the multicellular transgenic humanized C. elegans, and further validated the same in Mus musculus, besides testing their safety and toxicity. In our investigations, the final product—the HACBF (healthy ageing cognitive booster formulation) thus developed was found to reduce major aging biomarkers like lipofuscin, protein carbonyl, lipid levels and enhanced activity of antioxidant enzymes. Further confirmation was done using transgenic worms and RT-PCR. The cognitive boosting activities analyzed in C. elegans and M. musculus model system were found to be at par with donepezil and L-dopa, the two drugs which are commonly used to treat Parkinson’s and Alzheimer’s diseases. In the transgenic C. elegans model system, the HACBF exhibited reduced aggregation of misfolded disease proteins α-synuclein and increased the health of nicotinic acetylcholine receptor, levels of Acetylcholine and Dopamine contents respectively, the major neurotransmitters responsible for memory, language, learning behavior and movement. Molecular studies clearly indicate that HACBF upregulated major genes responsible for healthy aging and cognitive booster activities in C. elegans and as well as in M. musculus. As such, the present herbal product thus developed may be quite useful for healthy aging and cognitive boosting activities, and more so during this covid-19 pandemic.Graphical abstract
Economics of Vetiver Cultivation: Increase in the Income of Household from Marginal Land in Madhepura District of Bihar
The study was conducted in Madhepura district of Bihar to understand the socio-economic status of vetiver growers, profile of inputs used, relationship between cost and return, influence of input’s cost on yield and returns of vetiver. The study is absolutely based on primary information, collected from 70 farmers those who are benefited by CSIR-CIMAP Aroma Mission. The data were collected through field visit and adopted the personal interview method. The statistical tools like, descriptive analysis, CACP cost concept and multiple regression function were used to attain the objectives of the study. The results revealed that literacy rate of farmers in the study area was 82.42 percent whereas 96.80 percent of overall sample farmers were dependent on agriculture and 3.20 percent on Non-farm sources.
