Phage-antibiotic synergy (PAS) has been recently proposed as a superior strategy for the treatment of multidrug-resistant pathogens to effectively reduce bacterial load and slow down both phage and antibiotic resistance. However, the underlying mechanisms of resistance suppression by PAS have been poorly and rarely been studied. In this study, we tried to understand how PAS suppresses the emergence of resistance using a hypervirulent Klebsiella pneumoniae (KP) strain and a novel phage H5 in combination with ceftazidime (CAZ) as a model. Our study reveals a novel mechanism by which PAS drives altered evolutionary trajectory of bacterial populations, leading to suppressed emergence of resistance. The findings advance our understanding of how PAS suppresses the emergence of resistance, and are imperative for optimizing the efficacy of phage-antibiotic therapy to further improve clinical outcomes.

FBXO45 levels regulated ferroptosis renal tubular epithelial cells in a model of diabetic nephropathy by PLK1

This research aims to investigate the role and underlying biological mechanism of FBXO45 in regulating ferroptosis of renal fibrocytes in a diabetic nephropathy (DN) model.
C57BL/6 mice were fed with a high-fat diet and injected with streptozotocin to induce diabetes. Human renal glomerular endothelial cells stimulated with d-glucose.
Serum FBXO45 mRNA expression was found to be down-regulated in patients with DN. There was a negative correlation between the expression of serum FBXO45 mRNA and serum α-SMA, Collagen I, and E-cadherin mRNA in patients with DN. Additionally, the expression of serum FBXO45 mRNA showed a negative correlation with blood sugar levels. Based on a 3D model prediction, it was observed that FBXO45 interacts with polo-like kinase 1 (PLK1) at GLY-271, ILE-226, GLY-166, LEU-165, ARG-245, and ASN-220, while PLK1 interacts with FBXO45 at TYR-417, ARG-516, HIS-489, TYR-485, GLN-536, and ARG-557. This interaction was confirmed through immunoprecipitation assay, which showed the interlinking of FBXO45 protein with PLK1 protein.
These findings indicate that FBXO45 plays a role in mitigating ferroptosis in DN through the regulation of the PLK1/GPX4/SOX2 pathway. This highlights the potential of targeting FBXO45 as a therapeutic approach to ameliorate ferroptosis in DN.

Retrospective Analysis of Drug Sensitivity of Neisseria gonorrhoeae in Teaching Hospitals of South China

The aim of this study was retrospective analysis of drug sensitivity of Neisseria gonorrhoeae in two teaching hospitals of South China. A total of 304 Neisseria gonorrhoeae isolates obtained from patients in South China from 2016 to 2020 were evaluated. The MICs of penicillin, cefuroxime, ceftriaxone (CRO), cefepime, ciprofloxacin, ceftazidime and azithromycin (AZM) against the isolates were determined by the agar dilution method. Then, Neisseria gonorrhoeae isolates were categorized into sensitive, moderately sensitive and resistant according to MICs. Also, [beta]-lactamases were detected by enzyme linked immunosorbent assay (ELISA). Ureaplasma urealyticum and Mycoplasma hominis were determined by culture in liquid medium, and Chlamydia was detected by rapid antigen test. The result showed there was 50.99%, 20.72%, 9.87%, 14.47%, 86.84%, 7.57%, 6.91%, 11.18% resistance to penicillin, cefuroxime, ceftriaxone, cefepime, ciprofloxacin, ceftazidime and azithromycin, respectively. Also, [beta]-lactamase positivity was 53.29% and Chlamydia antigen positivity was 20.07%. Ureaplasma urealyticum and Mycoplasma hominis positivity was 11.84% and 6.25%, respectively. From 2016 to 2020, the resistant rate of ceftriaxone and azithromycin gradually increased. In conclusion, Southern China is among the area reporting gonococci with high-level resistance to AZM and CRO, so N. gonorrhoeae culture and drug sensitivity test will be vital for monitoring trends in antimicrobial resistance. Heat transfer performance of a porous copper micro-channel heat sink

A novel porous copper micro-channel heat sink based on the high thermal conductivity and structural stability of porous copper micro-channel material is proposed in this study. A circular cross-sectional and a multi-layer staggered arrangement are the main characteristics of the micro-channel heat sink. Additionally, a heat dissipation model is produced for porous copper micro-channel heat sink by analyzing the heat transfer process. Further, the theoretical heat transfer coefficient is calculated using MATLAB, and the experimental heat transfer coefficient is determined based on an experimental platform. The heat dissipation model is verified by comparing the theoretical and experimental heat transfer coefficients. The relation between the porous copper micro-channel heat sink parameters and the heat transfer coefficient that is obtained based on the heat dissipation model of the porous copper micro-channel heat sink is analyzed. The results exhibit that variations in pore diameter, porosity, porous copper length, porous copper width, porous copper height, and volume flow significantly affect the heat transfer performance.

自噬途径降解肝脏脂滴的研究进展

自噬是一种高度保守的细胞降解途径,可通过“脂噬”过程来降解脂滴。脂噬可以选择性地识别脂类物质并将其降解,促进β氧化,进而维持细胞内脂质代谢的平衡状态。肝脏通过脂噬信号通路或关键分子来调控脂滴代谢,进而降低肝脏脂肪变性,改善非酒精性脂肪性肝病。本文总结归纳了巨自噬、分子伴侣介导的自噬和微自噬样3种自噬途径降解肝脏脂滴的最新研究进展,AMPK/mTOR-ULK1、ATGL-SIRT1、FGF21-JMJD3、Akt作为调控脂噬过程的主要信号通路,有助于维持肝脂质代谢稳态,能够为临床预防和治疗非酒精性脂肪性肝病提供新思路。

基于相变材料GST与孔径共享宽带宽消色差透镜的设计

TN256; 提出了在超构透镜结构上采用孔径共享协同操作的方法,根据散单元输出相位与频率呈线性关系,再结合调控相变材料Ge2Sb2Te5 晶化率m值,在设定的波段内合理选择散射单元的尺寸,不同波长入射光经过超表面后能产生符合统一焦距要求的相位分布,从而使消色差超透镜的带宽得到相应拓宽,设计了9.5~13 μm连续波段内的偏振不敏感消色差超表面透镜.仿真结果表明,消色差超表面在工作波段内的焦距变化为3.57 μm,与设定焦距的误差约为4.3%,并且各波长入射下的焦点半峰全宽均达到了衍射极限,聚焦效率超过60%.提出的消色差超表面透镜,为消色差超表面的设计提供了新的思路,促进了相变材料在宽带消色差超表面方面的研究.

抑制性受体TIGIT与慢性HBV感染中免疫紊乱的关系

持续HBV感染改变了天然免疫细胞和获得性免疫细胞表面受体的表达,由此引发的多种免疫紊乱,可导致免疫逃逸,最终使疾病慢性化。研究表明,抑制性受体的上调是患者免疫紊乱的主要原因,阻断抑制性受体可一定程度上恢复患者的免疫功能。T淋巴细胞免疫球蛋白和免疫受体酪氨酸抑制性基序结构域(TIGIT)是目前较为关注的一种新型抑制性受体,在NK细胞和T淋巴细胞中高水平表达。研究发现,TIGIT在慢性病毒感染中发挥重要作用,现就TIGIT与慢性HBV感染中免疫紊乱的相关性研究进展进行简要综述。

驻南方某部军人反流性食管炎及幽门螺杆菌感染情况分析

目的研究南方某部军人反流性食管炎(RE)的患病情况及其与幽门螺杆菌(Hp)感染的关系。方法选择通过反流性疾病问卷(RDQ)调查诊断为胃食管反流病(GERD)的91例军人病例作为GERD组,并根据症状积分分为低分组(12~15分)47例,中分组(16~20分)34例,高分组(〉20分)10例。另随机抽取60例非GERD军人(反流症状评分〈12分)为对照组。共151例军人接受胃镜检查,同时采用14C-尿素呼气试验(14C-UBT)和快速尿素酶试验(RUT)两种方法检测Hp,结果均阳性者判为Hp感染。结果 GERD组与对照组的RE检出率分别为42.9%(39/91)和3.3%(2/60),两组比较差异有统计学意义(P〈0.05),但GERD各积分组之间的RE检出率差异无统计学意义(P〉0.05)。GERD组中检出39例RE,其中A、B级分别为221、3例(共占89.7%),C、D级分别为31、例(共占10.3%);另检出8例Barrett食管(BE,8.8%)。RE症状积分与分级之间无明显线性关系(r=0.276,P〉0.05)。RE患者Hp感染率为59.0%(23/39),与对照组(61.7%)比较差异无统计学意义(P〉0.05)。结论军人RE检出率及Hp感染率均较高,RE检出率与Hp感染无明显关联。

Acclimatization of Midrib Angle and Petiole Angle of Camptotheca acuminate Decne to Different Light Regimes in Evergreen Broadleaves Forests / 喜树幼苗的叶悬挂角和叶柄角对不同光环境的响应和适应

As a special deciduous broadleaf tree in evergreen broadleaves forests,Camptotheca acuminate is a class Ⅱ protected plant in China.In this paper,the light response and acclimation of leaf inclination in three different light regimes(open,gap and understory) was compared.The results indicated that significant difference among different regimes existed in ecological factors such as PAR,temperature,Vpd and etc.In different light regimes,the extents of diurnal change of midrib angle and petiole angle of Camptotheca acuminate were different,and the highest extent existed in open,the second was in forest gap,and the least was in understory.The midrib angle and the petiole angle appeared at \"peak\" at 16:00,14:00 and 12:00 respectively in these three light regimes in the sunny day in summer,correspondingly,the values of midrib angle were about 140°,128° and 112°,and the values of petiole angle were about 67°,59° and 55° respectively.In the sunny day,diurnal courses of midrib angle and petiole angle of Camptotheca acu
