Lack of vascularization is directly associated with refractory wound healing in diabetes mellitus (DM). Enrichment of endothelial precursor cells (EPCs) is a promising but challenging approach for the treatment of diabetic wounds. Herein, we investigate the action of nicotinamide riboside (NR) on EPC function for improved healing of diabetic wounds. Db/db mice that were treated with NR-supplemented food (400 mg/kg/d) for 12 weeks exhibited higher wound healing rates and angiogenesis than untreated db/db mice. In agreement with this phenotype, NR supplementation significantly increased the number of blood EPCs and bone marrow (BM)-derived EPCs of db/db mice, as well as the tube formation and adhesion functions of BM-EPCs. Furthermore, NR-supplemented BM-EPCs showed higher expression of sirtuin 1 (Sirt1), phosphorylated adenosine monophosphate–activated protein kinase (p-AMPK), and lower expression of acetylated peroxisome proliferator–activated receptor γ coactivator (PGC-1α) than BM-EPCs isolated from untreated db/db mice. Knockdown of Sirt1 in BM-EPCs significantly abolished the tube formation and adhesion function of NR as well as the expression of p-AMPK and deacetylated PGC-1a. Inhibition of AMPK abolished the NR-regulated EPC function but had no effect on Sirt1 expression, demonstrating that NR enhances EPC function through the Sirt1-AMPK pathway. Overall, this study demonstrates that the oral uptake of NR enhances the EPC function to promote diabetic wound healing, indicating that NR supplementation might be a promising strategy to prevent the progression of diabetic complications.

Experiences of working as a clinical nurse while pregnant during the coronavirus disease-2019 pandemic: a qualitative study

Background
Working as a pregnant clinical nurse might experience a range of challenges, such as significant anatomical and physiological changes as well as emotional and cognitive changes. That might be particularly obvious under the historical background of coronavirus disease-2019 (COVID-19) pandemic. However, a dearth of studies has explored the experiences of working as a pregnant nurse during this special period. This study aimed to explore the experiences of working as a clinical nurse while pregnant during the COVID-19 pandemic.
Methods
A descriptive qualitative design was selected. Purposive sampling, combined with maximum variation strategy and snowball sampling, were utilized to identify and select participants from tertiary-teaching hospitals, specialized hospitals, and community hospitals in Zhejiang Province, southeastern China. Online semi-structured individual interviews were used to collect data, and conventional content analysis was used to analyze the data.
Results
Eleven Chinese nurses with a mean age of 31.8 years, ranging from 26 to 40 years, participated in this study. Four themes and twelve subthemes emerged: (1) still adhering to work as a clinical nurse despite being pregnant during the pandemic; (2) working during pregnancy under pandemic is still an ordinary nurse; (3) still staying in the special life phase as a pregnant mother; and (4) growth and gains as pregnant mother.
Conclusion
The pregnant clinical nurses suffered from various changes and difficulties during the pandemic. Managers, occupational health and other health system leaders, and policymakers should be aware of the importance of establishing a work environment that guarantees safe continued pregnancy. Future studies should focus on the establishment of specific guidelines and manuals regarding how pregnant nurses worked, as well as the development of self-protection interventions during pregnancy. Moreover, research on moral stigma and bullying in nursing during pregnancy deserves further exploration.

Perceptions and experiences of a multicomponent traditional Chinese medicine lifestyle medicine program for depression: a qualitative study
Background
Depression, a debilitating mental disorder, has become the leading cause of disability worldwide. A growing body of evidence supports the feasibility and effectiveness of multicomponent lifestyle medicine programs for the treatment of depression, including the recent novel multicomponent Traditional Chinese Medicine Lifestyle Medicine (TCMLM) program (Registered at ClinicalTrials.Gov with registration number: NCT05799586). However, little is known about participants’ experiences and perceptions of the program and the aspects that require improvement. This study aimed to explore the experiences and perceptions of participants attending the multicomponent TCMLM program and practicing the related lifestyle behaviors.
Methods
In this descriptive qualitative study, purposeful sampling was used to recruit Hong Kong Chinese adults who had participated in the multicomponent TCMLM program between August 2023 and January 2024. Face-to-face focus-group interviews and semi-structured interviews were conducted with audio recording, transcribed verbatim, and analyzed using conventional content analysis. All interviews were performed in classrooms in a university in Hong Kong.
Results
A total of 31 multicomponent TCMLM program attendees aged 20 to 65 years participated in the qualitative interviews. The content analysis identified three themes and 12 subthemes, namely, Theme 1: multicomponent TCMLM program originally being comprehensive and special (e.g., TCMLM program content originally being not unitary, increasing the number of methods for dealing with depressive symptoms); Theme 2: multicomponent TCMLM program being far more beneficial than expected (e.g., promoting diet, exercises, daily routine and sleep management based on TCMLM, improving physical functional status, improving one’s personality); and Theme 3: practicing multicomponent TCMLM program having challenges (e.g., unsuitable conditions hindering some TCMLM practices in community).
Conclusions
This study provides fresh in-depth insights into the perceptions and experiences of Hong Kong Chinese adults with depression who attended the multicomponent TCMLM program and engaged in the related lifestyle behaviors. Meanwhile, the challenges encountered while attending the program and practicing the related behaviors offer valuable information for further optimization of the program and expanding its application in Hong Kong or other regions.

Adaptation Computing Parameters of Pan-Tilt-Zoom Cameras for Traffic Monitoring

The Closed-CIRCUIT television (CCTV) cameras have been widely used in recent years for traffic monitoring and surveillance applications. The researchers can use CCTV cameras to extract automatically real-time traffic parameters according to the image processing and tracking technologies. Especially, the pan-tilt-zoom (PTZ) cameras can provide flexible view selection as well as a wider observation range, and this makes the traffic parameters can be accurately calculated. Therefore, that the parameters of PTZ cameras are calibrated plays an important role in vision-based traffic applications. However, in the specific traffic environment, which is that the license plate number of the illegal parking is located, the parameters of PTZ cameras have to be updated according to the position and distance of illegal parking. In proposed traffic monitoring systems, they use the ordinary webcam and PTZ camera. They get vanishing-point of traffic lane lines in the pixel-based coordinate system by fixed webcam. The parameters of PTZ camera can be initialized by distance of the traffic monitoring and specific objectives and vanishing-point.

Miniemulsion cross-linking: A convenient route to hollow polymeric nanocapsule with a liquid core

Miniemulsion stabilized by poly(2-(dimethylamino) ethyl methacrylate)-block-poly(butyl methylacrylate) (PDMAEMA-b-PBMA) diblock copolymers has been used as liquid templates for the synthesis of polymer nanocapsules via quaternization cross-linking of PDMAEMA segments of the copolymer by 1,2-bis(2-iodoethoxy)ethane (BIEE) crosslinkers. PDMAEMA-b-PBMAs here as a stabilizer in miniemulsion with different molecular weights led to a size variation in diameters of nanocapsules, demonstrating the capsules have potential design capability of this technique. The solution behavior of the capsules has been also investigated in this paper.

Analysis of the Group Characteristics of Yuhua Heroes based on Lightweight Ontology

[Purpose/Significance] The spirit of Yuhuatai martyrs is one of the important components of the new democratic revolutionary spirit of the Communist Party of China (CPC). The primary documents about Yuhuatai martyrs is the significant source to record their spirit. Using text processing technologies, such as content annotation and feature construction to deeply excavate the content of the documents, which is not only conducive to carrying forward revolutionary culture and continuing the red spirit, but also beneficial to expanding the application objects of technology and promoting the improvement of text mining technology. [Method/Process] In this paper, the original documents related to Yuhuatai martyrs were used as the data source, which include 12,329 original records in 8 types of documents, including biographies of martyrs, letters, diaries, interrogation materials of the enemy and puppet governments, trial records, and oral history. Then the lightweight ontology of Yuhuatai martyrs was built, which was used as the feature description framework of Yuhuatai martyrs. Based on the domain ontology and trigger word features, relevant instances in the ontology were extracted from the documents of Yuhuatai martyrs, and the formal features of the multi-dimensional attributes of Yuhuatai martyrs were constructed and described. The categories and attributes of lightweight ontology described the Yuhuatai martyrs' group feature framework. Through the data analysis of the extracted ontology instance, the data analysis of Yuhuatai martyr group feature can be realized. [Results/Conclusions] Through the statistical analysis of the basic information, revolutionary activities and the relevant text content of the struggle in prison of Yuhuatai martyrs, this paper further revealed the group characteristics from the perspective of data, including: high education level, sacrifice at a young age, long-term struggle in the ruling center of the Kuomintang, adherence to faith, loyalty to CPC, full of family and country feelings of peace and order, and conscious practice of noble moral pursuit. By the application of lightweight ontology and other quantitative analysis methods in this paper, it is helpful to visualize the lofty ideals and beliefs of Yuhuatai martyrs, noble moral sentiments and the dauntless spirit of sacrifice for the people. This paper has a certain reference value for the presentation and dissemination of innovative Yuhuatai martyrs' spirit.

Initiator-free photocrosslinking of electrospun biodegradable polyester fiber based tubular scaffolds and their cell affinity for vascular tissue engineering

Random copolyester of poly(ɛ-caprolactone-co-L-lactide) (PCLA) with a 50:50 feeding molar ratio was synthesized via the ring-opening polymerization and functionalized by the end-capping reaction with acryloyl chloride. The resulting acrylated PCLA was then fabricated into small diameter tubular scaffolds by electrospinning technique and the formed scaffolds were followed by photocrosslinking under UV irradiation in the absence of photoinitiator. The mechanical strengths including tensile, suture retention and burst pressure were greatly enhanced after the photocrosslinking. The in vitro degradation data clearly revealed that the mechanical properties of the crosslinked scaffolds still remained after one month degradation in PBS solution, while those of the non-crosslinked ones lost heavily. The cytotoxicity assay on the mouse fibroblast L929 cells was conducted via MTT measurement. Furthermore, the observation on endothelial and fibroblast cell adhesion and proliferation was also made by using scanning electron microscopy (SEM). The initiator-free photocrosslinked tubular scaffolds show the potential to be used in vascular tissue engineering.

Insulin-stimulated glucose uptake by the glucose transporter GLUT4 plays a central role in whole-body glucose homeostasis, dysregulation of which leads to type 2 diabetes. However, the molecular components and mechanisms regulating insulin-stimulated glucose uptake remain largely unclear. Here, we demonstrate that Axin interacts with the ADP-ribosylase tankyrase 2 (TNKS2) and the kinesin motor protein KIF3A, forming a ternary complex crucial for GLUT4 transiocation in response to insulin. Specific knockdown of the individual components of the complex attenuated insulin-stimulated GLUT4 translocation to the plasma membrane. Importantly, TNKS2-/- mice exhibit reduced insulin sensitivity and higher blood glucose levels when re-fed after fasting. Mechanistically, we demonstrate that in the absence of insulin, Axin, TNKS and KIF3A are co-localized with GLUT4 on the trans-Goigi network. Insulin treatment suppresses the ADP-ribosylase activity of TNKS, leading to a reduction in ADP ribosylation and ubiquitination of both Axin and TNKS, and a concurrent stabilization of the complex. Inhibition of Akt, the major effector kinase of insulin signaling, abrogates the insulin-mediated complex stabilization. We have thus elucidated a new protein complex that is directly associated with the motor protein kinesin in insulin-stimulated GLUT4 translo- cation.

硫氧还蛋白还原酶抑制剂乙烷硒啉逆转顺铂耐药及其机制研究

目的:研究凋亡调控相关蛋白来了解顺铂耐药成因,同时考察乙烷硒啉(Ethaselen)在K562耐药细胞中逆转顺铂耐药的作用,并初步探讨其作用机制。创新点:首次研究乙烷硒啉在逆转顺铂耐药中的作用,且此作用与乙烷硒啉诱导细胞凋亡相关。方法:通过长时间脉冲诱导得到顺铂耐药K562细胞,并观察耐药细胞形态及倍增时间。采用MTT法考察乙烷硒啉、顺铂及其联用组在不同细胞株间的生长抑制作用。流式细胞术分析细胞凋亡情况以及细胞内活性氧(ROS)水平。最后,通过蛋白质免疫印迹(Western blot)考察凋亡调控相关蛋白水平的变化。结论:脉冲诱导得到的K562耐药细胞对顺铂的耐受性是原K562细胞的5.34倍。形态学观察发现,耐药细胞体积增大,粘附性进一步降低。乙烷硒啉与顺铂联用表现出协同效应。当加入少量的乙烷硒啉(顺铂与乙烷硒啉的摩尔比率为10:1),顺铂作用K562耐药细胞的半抑制浓度(IC50)值可以减少21倍。流式细胞术及Western blot表明,乙烷硒啉能够诱导耐药细胞凋亡。其逆转顺铂耐药主要是通过调控Bcl-2及Bax蛋白比例以及通过提高细胞内活性氧水平引起线粒体通透转运孔道(PTP)蛋白孔道的形成来促使释放细胞色素c,进而引起Caspase凋亡途径。
