Circular RNA hsa_circ_0000730 restrains cell proliferation, migration, and invasion in cervical cancer through miR‐942‐5p/PTEN axis

Circular RNAs (circRNAs) play prominent roles in regulating the progression of cancers. This study is aimed to decipher the role of hsa_circ_0000730 in cervical cancer (CC).The differentially expressed circRNAs of CC were screened out from the Gene Expression Omnibus database. qRT‐PCR was used to detect circ_0000730 expression in CC tissues and cell lines, and the Kaplan–Meier curve was adopted to figure out the relationship between circ_000730 expression and the overall survival time of CC patients. BrdU assay and Tanswell assay were utilized to examine the proliferation, migration, and invasion of CC cells. Western blot was adopted to detect PTEN protein expression. Bioinformatics analysis and dual‐luciferase reporter assay were used to examine the target relationship between miR‐942‐5p and circ_0000730 or PTEN, respectively.Circ_0000730 was among the differentially expressed circRNAs in CC. Circ_0000730 was significantly down‐regulated in the cancer tissues of 50 CC patients and CC cell lines. Additionally, underexpression of circ_0000730 was associated with the shorter survival time of CC patients. Gain‐ and loss‐of‐function assays highlighted that circ_0000730 significantly inhibited the proliferation, migration, and invasion of CC cells. Mechanistically, miR‐942‐5p was identified as a downstream target of circ_0000730, and circ_0000730 could positively regulate PTEN expression via repressing miR‐942‐5p in CC cells.Circ_0000730 inhibits the proliferation, migration, and invasion of CC cells via regulating miR‐942‐5p/PTEN axis. Circ_0000730 probably acts as a tumor suppressor in CC, and it may be a candidate target for the treatment of CC.

Gap Resonance Between a Stationary Box and a Vertical Wall Induced by Transient Focused Wave Groups

This paper explores the phenomenon of fluid resonance occurring within a narrow gap between a vessel and a vertical wharf, taking ships berthing in front of a gravity wharf as the research background. Using the open-source software OpenFOAM®, a two-dimensional viscous-flow numerical wave flume was developed to simulate the fluid resonant motions induced by transient focused wave groups with different spectral peak periods and wave amplitudes. The results indicate that for all the incident focused wave amplitudes considered, the amplitudes of the free surface elevation in the gap, horizontal wave force and moment all exhibit a bimodal variation trend with increasing spectral peak period. The peak values of the above amplitude-period curve appear near the resonant period of the first and second harmonic components of the free surface elevation. However, the variation in the vertical wave force versus the spectral peak period presents different patterns. In addition, the first- to fourth-order harmonic components in the wave surface and forces are further examined via the four-phase combination method. The results show that the first- to second-order harmonic components play a dominant role in the overall amplitude.

Notch1-ADAM8 positive feed-back loop regulates the degradation of chondrogenic extracellular matrix and osteoarthritis progression

Background
Osteoarthritis (OA) is one of the most prevalent joint disease, and there are still no effective therapeutic agents or clinical methods for the cure of this disease to date. The degradation of cartilage extracellular matrix (ECM) is a major cause of OA.
Method
IL-1β was used to induce chondrogenic degradation. Q-PCR and Western blotting were used to detect mRNA and protein level, respectively. ELISA was used to detect the secreted TNF-α and IL-6 level. Immunofluorescence was used to detect the protein level of Aggrecan, Collagen II and ki67. TUNEL and flow cytometry were used to examine cell apoptosis of chondrocytes. ChIP and luciferase assay were used to study molecular gene regulation. Osteoarthritic animal model and Safranin-O staining were used to determine the in vivo OA phenotype.
Results
The expression of ADAM8 was up-regulated in osteoarthritic chondrocytes. Knockdown of ADAM8 suppressed the OA phenotype in the in vitro OA cell model. ADAM8 regulated OA progression through the activation of EGFR/ERK/NF-κB signaling pathway. Inhibition of Notch signaling suppressed OA phenotype in the in vitro OA cell model. Notch signaling regulated the gene expression of ADAM8 directly via Hes1. Notch1-ADAM8 positive feedback loop promoted the progression of OA in vivo.
Conclusion
Notch1-ADAM8 feed-back loop regulates the degradation of chondrogenic extracellular matrix and osteoarthritis progression.

High spontaneous male-fertility-restorer frequency in a maize recurrent selection experiment

Using the crosses of the DH lines derived from a single cross hybrid Xianyu 335 as source materials for haploid induction in the recurrent selection scheme, we found that the rate of spontaneous chromosome doubling in the tassels of the haploid plants increased significantly in the second cycle and showed a cumulative genetic effect, and through using DH recurrent selection technique, the doubling rate of the haploid tassels can be doubled. Proved performing continuous recurrent selection in a narrow single cross background, the genetic makeup of the haploids had been enriched for spontaneous doubling restoration. Using hybrids between DH lines from two cycles haploid selection as source materials for haploid induction, the haploid tassel doubling rate had reached 85.15% and haploid ears showed seed set had reached 66.18%, representing a rate increase of 6.99 folds and 9.86 folds of its original source population Xianyu335.The genetic gain of set seeds plant frequency is 29.74% per cycle. Also, because the DH lines had been selected from two cycles of haploids or whole genome gamete selection based on the haploid phenotypes, it was effectively eliminated many deleterious and bad genes, and also cumulatively combined many superior genes for agronomy and plant growth vigor. These new DHs can be directly used as parents for germplasm enhancement and new hybrid selection and breeding. The discovery of the enrichment for spontaneous chromosome doubling and fertility restoration in of the tassels of the haploid plants tassels that were derived from DH recurrent selection technique and create high spontaneous doubling rate source materials may have showed a significant application value in maize breeding. It shows significant advantages for construct maize two cycle. Recurrent selection with high haploid spontaneous doubling rate can to speed up breeding output and cumulative genetic improvements.

Effective treatment of apatinib in desmoplastic small round cell tumor: a case report and literature review

Background
Desmoplastic small round cell tumor (DSRCT) is a rare malignant sarcoma with poor prognosis due to lack of effective treatments. Apatinib is a new potent oral small-molecule tyrosine kinase inhibitor, and targets the intracellular domain of vascular endothelial growth factor receptor 2 (VEGFR-2). In this study, we presented a case of intra-abdominal DSRCT which was effectively treated by apatinib.
Case presentation
A 32-year-old man was admitted due to increasing urination frequency and palpable mass in right lower abdomen for 2 months. The mass was resected and diagnosed DSRCT. The patient refused chemotherapy and radiotherapy,and used Chinese medicine only. Six months after the surgery, the patient re-hospitalized due to growing abdominal mass and ascites. Intraperitoneal cisplatin treatment showed little effect. Apatinib was then recommended. Apatinib revealed outstanding effect on reducing mass size and ascites during 2-month treatment. Apatinib therapy continued for additional 2 months, and the patient was in good condition. The only toxicity was hand-food syndrome, which was controllable and well tolerated.
Conclusion
It is the first report that apatinib is effective on DSRCT. This report may provide an additional option for the treatment of metastatic DSRCT.

Overexpression of GLP-1 receptors suppresses proliferation and cytokine release by airway smooth muscle cells of patients with chronic obstructive pulmonary disease via activation of ABCA1

Glucagon-like peptide-1 (GLP-1) is an important insulin secretagogue that possesses anti-inflammatory effects. GLP-1 receptor (GLP-1R) agonists have been demonstrated to serve a pivotal role in the treatment of obstructive lung diseases, including chronic obstructive pulmonary disease (COPD). However, the specific function and underlying mechanisms of GLP-1R in COPD remain uncertain. The aim of the present study was to investigate the action and underlying mechanisms of GLP-1R in airway smooth muscle (ASM) cells from COPD patients. GLP-1R expression levels were markedly decreased in ASM cells from COPD patients compared with those from healthy controls. ASM cell proliferation and migration, and the levels of the inflammatory cytokines interleukin (IL)-1β, IL-4, tumor necrosis factor (TNF)-α, and granulocyte-macrophage colony-stimulating factor (GM-CSF) were measured. Transfection of pcDNA3.1-GLP-1R had inhibitory effects on ASM cell proliferation and migration, whereas GLP-1R small interfering (si)RNA reversed these effects. Furthermore, the present study demonstrated that GLP-1R overexpression markedly suppressed IL-1β, IL-4, TNF-α and GM-CSF levels. GLP-1R overexpression upregulated the expression levels of adenosine triphosphate-binding cassette, subfamily A, member 1 (ABCA1) in ASM cells, and the effects of GLP-1R on cell proliferation and migration, and inflammatory cytokine expression in ASM cells was abolished by siRNA-mediated silencing of ABCA1. The results of the present study suggested that GLP-1R contributes to COPD pathology, potentially via an ABCA1-mediated pathway.

This paper combines the global optimization ability of the symbiotic parallel genetic algorithm and the local optimization ability of the improved LMBP algorithm to research,proposes an neural network structure optimization symbiotic parallel genetic algorithm and to testify the correctness and validity of this algorithm by the simulation experiments. This algorithm realizes unequal length coding, large probability cross, small probability variation, cross and variation between sub-populations, information exchanging between sub-populations etc, and successful implements the optimization of neural network structure. The experimental results shows that this algorithm having reliable performance, searching a large space, be able to find the feasible solution within the specified generalization and approximation error range.

In view of commercialization, however, the maximum superconducting currents for coated conductors should be increased further. Unfortunately, it has been frequently observed that the average critical current density

decreases with an increase in film thickness. The thickness effect is still a hurdle for large-scale production, especially in pulsed laser deposition and metal organic deposition processes. An engineering current of more than 1 000 A/cm is desired owing to the high cost of 2G superconducting materials. The present work attempts to review the evolution of various issues subject to the thickness effect, including the microstructure, epitaxial texture, surface roughness, pinning force, oxygen deficiency, residual stress, copper-rich layers, and segregation of elements. Furthermore, recent progress in enhancing the performance of superconductors especially in terms of critical current density is illustrated, such as the use of heavy doping. Further understanding of the thickness effect is extremely important for large-scale commercial development of the second generation high temperature superconductors.

Analysis on temporal and spatial variations of groundwater hydrochemical characteristics in the past decade in southern plain of Beijing, China
Groundwater utilization and protection are crucial for sustainable urban development. This is especially true for Beijing, where groundwater is an important source for urban water supply. In this study, statistical methods, including descriptive statistics, correlation analysis, principal component analysis, and Piper-Tri-linear diagram, were used in analyzing the temporal and spatial variations of the hydrochemical characteristics of groundwater based on monitored data from the southern plain of Beijing, China. Results indicated consistent changes of groundwater's hydrochemical characteristics in different aquifers in the study area. The percentage of HCO3- in total anion increased significantly in the groundwater, and hydrochemical water type evolved gradually from Ca-Mg-CI-SO4 based to Ca-Mg-HCO3 based from period 2005-2007 to period 2013-2015. In shallow groundwater, the concentration of Na+, Ca2+, SO42, HCO3-, and total dissolved solids (TDS) increased from period 2005-2007 to period 2013-2015, and the greatest change came from HCO3@, rising from 428.93 to 528.96 mgL-1. The changes of main ionic concentrations in the deep groundwater were consistent with those in the shallow groundwater for both periods. However, the variations in deep groundwater were less than those in shallow groundwater. The temporal and spatial variations of hydrochemical characteristics reflect the groundwater quality in the study area. This study could facilitate decision-making process on the protection of groundwater resources to ensure its sustainable utilization.
