To determine the efficacy and safety profile of rhubarb and α-keto analogs of
essential amino acids supplementation in patients of diabetic nephropathy (DN), we studied 96
patients of DN attending a tertiary care center of the North India. The patients were randomly
divided into three equal interventional groups. Group I (control) that received conservative
management along with placebo, Group II (rhubarb) that received conservative management
along with rhubarb capsule (350 mg, thrice daily), and Group III [keto amino acid (KAA)] that
received conservative management along with α-keto analogs of essential amino acids (600 mg,
thrice daily). The treatment was continued for 12 weeks. Clinical and biochemical parameters
were assessed at 0, 4, 8, and 12 weeks of treatment. A progressive improvement in clinical features
and biochemical parameters was seen in all three groups after 12 weeks of treatment. The
KAA group showed more marked improvement in clinical features as well as biochemical parameters
compared to the rhubarb group. There was a reduction in blood glucose, blood urea, serum
creatinine, and 24 h total urine protein. There was an increase in hemoglobin, 24 h total urine
volume, and glomerular filtration rate. There was no statistical difference between the rhubarb
and KAA groups with respect to side effects (P > 0.05). Our study suggests that KAA is more
effective than rhubarb as add-on therapy with conservative management in patients of DN.

Stability constants of mixed ligand complexes of lead(II) with 1-(aminomethyl) cyclohexane acetic acid and α-amino acids

Formation of binary and ternary complexes of lead(II) ions with 1-(aminomethyl) cyclohexane acetic acid and some biologically important α-amino acids, such as glycine, l-alanine, l-valine, l-leucine, l-isoleucine, l-phenylalanine and l-proline was investigated using the potentiometric technique at 32 °C. The properties of mixed ligands were investigated and discussed. The acidity constants of the ligands and their stability constants were determined in 50% (v/v) DMSO-water medium under experimental conditions. The ternary complex formation was found to occur in a stepwise manner. The stability of ternary complexes was investigated and compared with that of the corresponding binary complex in terms of the parameters, Δ log K and log X. The concentration distribution of various species formed in the mixed ligand systems was evaluated.

Herein we provide a detailed result about the effect of various additives, viz. inorganic salts, quaternary ammonium bromides (QABs) and amino acids on clouding behavior of amphiphilic drug amitriptyline hydrochloride (AMT). The continuous increase in the cloud point (CP) of drug by increase in inorganic salt concentration and the magnitude of increases rely upon the position of the salts in Hofmeister series and hydrated radii. The QABs also influence continuous increase in the CP, which is illustrated in terms of the alkyl chain length of peculiar QAB. The effect of amino acids on CP of the drug solution is dependent upon the characteristics (acidic, basic, polar or nonpolar) of particular amino acids. The overall behavior of additives has been analyzed and discussed on the basis of electrostatic repulsion or interaction, micellar growth, and mixed micelle formation between the ingredients. In addition to this, thermodynamic parameters are also evaluated.

The protein selectivity index as measured from the ratio of urinary immunoglobulin
to albumin failed to differentiate between steroid-sensitive (SS) and steroid-resistant
(SR) cases of nephrotic syndrome (NS). Sialic acid contributes negative charges to many plasma
proteins. The negative charge is a determinant of protein excretion rate. The prognostic significance
of assay of urinary excretion of protein-bound sialic acid in NS has not been evaluated.
Hence, the present study was designed to evaluate whether measurement of urinary protein -
bound sialic acid (UPBSA) can be used as a marker to differentiate SS from SR cases of NS. The
urine samples of 70 (47 SS and 23 SR) pediatric NS children were assayed for UPBSA by
Aminoff’s method. The levels were compared and the receiver–operator curve was drawn to
determine the optimum cutoff point to differentiate among the groups before starting the therapy.
The excretion of UPBSA in SR cases of NS was significantly higher than that of SS cases (P
<0.05). The optimum cutoff limit for UPBSA was 2.71 μg/mg of proteins with 75% sensitivity
and 75.5% specificity for differentiating SS cases from SR cases (area under the plasma–
concentration time curve = 0.814, P = 0.009). We conclude that UPBSA can differentiate SR
cases from SS cases of NS in pediatric patients and may help in predicting the response to steroid
therapy.

Effect of chemical compounds on amino acid content of some Fusarium species and its significance to fungal chemotaxonomy

The cellular amino acid profiles of nine species of Fusarium; namely, Fusarium anthophilum, Fusarium avenaceum, Fusarium cerealis, Fusarium graminearum, Fusarium graminum, Fusarium oxysporum f. sp. conglutinans, Fusarium pseudograminearum, Fusarium roseum, and Fusariumsacchari var. elongatum growing on malt extract medium were determined. The amino acid profiles of the investigated fungi were varied and could be used for identification and characterisation of certain Fusarium species. Addition of certain chemical compounds including aspartic acid, glutamic acid, methionine, selenium, and urea to the growth medium affected the amino acid profiles. However, susceptibility of amino acid content to environmental conditions increased the variation of amino acid profiles among all the investigated Fusarium species. Some amino acids were only produced when certain chemical compounds were added to the growth medium. Valine was produced by F. anthophilum only in the presence of aspartic acid or selenium, while serine was produced in the presence of aspartic acid, glutamic acid, or methionine. Also, cysteine was produced by F. avenaceum in the presence of glutamic acid or urea. F. cerealis produced tryptophan only in the presence of aspartic acid or urea, while F. graminearum produced leucine in glutamic, methionine or urea. Similarly, many different amino acids were produced by each Fusarium species only in the presence of certain chemical compounds. The results revealed that the amino acid profiles will be more useful for characterisation and identification of fungi if they are determined under different conditions.
