Atypical myopathy: an update

Atypical myopathy is an acute intoxication induced by the ingestion of hypoglycin A, a toxin contained in the seeds and seedlings of certain trees of the genus Acer (particularly Acer pseudoplatanus [sycamore] in Europe), which results in disruption of the energy metabolism. The clinical signs arise from energy depletion in the type I fibres of the postural and respiratory muscles, and lead to a specific biochemical profile that may be used to confirm a diagnosis. Since there is no cure for the condition, the emphasis is on prevention, which requires knowledge of the conditions necessary for the causative agent to exert its toxicity. This article gives an overview of atypical myopathy, discussing the mechanism involved, its aetiology and the clinical signs and management (therapeutic and prevention).

To identify novel antiapoptotic proteins encoded by DNA viruses, we searched viral genomes for proteins that might interfere with Fas and TNFR1 apoptotic signaling pathways. We report here that equine herpesvirus type 2 E8 protein and molluscum contagiosum virus MC159 protein both show sequence similarity to the death effector domains (DEDs) of the Fas/TNFR1 signaling components FADD and caspase-8. Yeast two-hybrid analysis revealed that E8 protein interacted with the caspase-8 prodomain whereas MC159 protein interacted with FADD. Furthermore, expression of either E8 protein or MC159 protein protected cells from Fas- and TNFR1-induced apoptosis indicating that certain herpes-viruses and poxviruses use DED-mediated interactions to interfere with apoptotic signaling pathways. These findings identify a novel control point exploited by viruses to regulate Fas- and TNFR1-mediated apoptosis

Hypoglycin A is a plant-derived protoxin that causes atypical myopathy in equids. In atypical myopathy-affected horses, metabolomic and microbiome studies have reported alterations in metabolic markers and faecal microbiota composition, pointing to a potential disruption of microbial homeostasis. However, in vivo observations are strongly confounded by host-related factors, underscoring the need for controlled in vitro approaches. To address this, we used an in vitro static batch fermentation model simulating the equine colon to investigate the direct effects of hypoglycin A on microbiota composition and activity. Faecal inocula from healthy horses were incubated in control and hypoglycin A-treated fermenters for 48 h, with serial analyses of hypoglycin A concentration, short-chain fatty acids, and 16S rRNA gene profiles. Hypoglycin A remained stable in the nutritive medium in the absence of microbiota, confirming that its degradation in inoculated fermenters was microbiota-dependent. The results showed significant microbial-associated hypoglycin A degradation without evidence of toxic metabolite formation. The analysis of α- and β-diversity revealed both an effect of incubation time, reflecting the natural temporal dynamics of microbial communities under batch fermentation, and a specific impact of hypoglycin A exposure, with certain taxa such as Paraclostridium being affected. This study provides the first in vitro evidence that the equine microbiota contributes to hypoglycin A degradation.

Hypoglycin A and methylenecyclopropylglycine are protoxins responsible for atypical myopathy in equids. These protoxins are converted into toxins that inhibit fatty acid β-oxidation, leading to blood accumulation of acylcarnitines and toxin conjugates, such as methylenecyclopropylacetyl-carnitine. The enzymes involved in this activation are also present in some prokaryotic cells, raising questions about the potential role of intestinal microbiota in the development of intoxication. Differences have been noted between the faecal microbiota of cograzers and atypical myopathy-affected horses. However, recent blood acylcarnitines profiling revealed subclinical cases among cograzers, challenging their status as a control group. This study investigates the faecal microbiota of horses clinically affected by atypical myopathy, their cograzers, and a control group of toxin-free horses while analysing correlations between microbiota composition and blood parameters. Faecal samples were analysed using 16S amplicon sequencing, revealing significant differences in α-diversity, evenness, and β-diversity. Notable differences were found between several genera, especially Clostridia_ge, Bacteria_ge, Firmicutes_ge, Fibrobacter, and NK4A214_group. Blood levels of methylenecyclopropylacetyl-carnitine and C14:1 correlated with variations in faecal microbial composition. The theoretical presence of enzymes in bacterial populations was also investigated. These results underscore the critical need to investigate the potential role of intestinal microbiota in this poisoning and may provide insights for developing prevention and treatment strategies.

Le pied bot varus équin (PBVE) invétéré est une attitude vicieuse du pied qui ne repose plus sur le sol par ses points d'appui normaux mais par son bord externe et sa pointe. Lorsque cette attitude vicieuse persiste longtemps, elle évolue vers des déformations ostéoarticulaires importantes à l'âge adulte. Le PBVE fixé constitue une bonne indication d'arthrodèse tibio-calcanéenne. Celle-ci permet dans la majorité des cas d'obtenir un appui plantigrade indolore et stable. Le but de ce travail est d'évaluer les résultats cliniques et radiologiques de l'astragalectomie avec adhèse tibiocalcanéenne. Nous rapportons une série de 3 cas d'arthrodèse tibio-calcanéenne pour déformation grave de l'arrière-pied associant un varus et un équin important et gênant chez l'adulte. Les résultats étaient jugés satisfaisants avec un appui plantigrade indolore et stable. Avec fusion de l'arthrodèse à 90 jours, sans inégalité pour le cas bilatéral et un raccourcissement de 2cm compensé par une semelle à droite pour le cas unilatéral. Dans les déformations importantes et fixées de la cheville et du PBVE, l'arthrodèse tibio-calcanéenne permet dans la majorité des cas d'obtenir un appui plantigrade indolore et stable sans risque vasculaire, ni cutané.

La place de la double arthrodèse dans la prise en charge du pied bot varus équin invétéré

La prise en charge du pied bot varus équin invétéré ( PBVEI) pose d’énormes problèmes thérapeutiques. La double arthrodèse sous-talienne et médio-tarsienne longtemps considérée comme la solution de sécurité pour ces déformations est encore couramment utilisée. Nous rapportons une série de 13 enfants opérés pour un pied bot varus équin invétérés (16 pieds) par Arthrodèse sous-talienne et médio tarsienne réalisée au service d'orthopédie pédiatrique du CHU Hassan II; de Fès au Maroc sur une période de 4 ans; étalée de janvier 2009 à décembre 2012. L’âge moyen de nos patients était de 12,6 ans avec prédominance féminine. L'origine congénitale était retrouvée chez 10 patients. L'atteinte était gauche chez 8 patients avec une localisation bilatérale chez 3 patients. La radiographie standard du pied de face et de profil a révélée une divergence talo-calcanéenne qui variait entre 5 et 20°, l'angle talus-1er métatarsien entre 20 et 40° (avec une moyenne de 28°) et l'angle calcanéus-5ème métatarsien entre 15° et 45° (avec une moyenne de 30°). Tous les patients ont bénéficiés d'une arthrodèse sous-talienne et médio tarsienne. Les résultats étaient satisfaisants dans 98% des cas. Le pied était plantigrade dans 9 cas, le varus de l'arrière pied persistait dans 4 pieds alors que l’équin et le varus de l'avant pied étaient notés chez 2 cas. La double arthrodèse est l'intervention idéale pour stabiliser et corriger les déformations rencontrées dans le PBVE invétéré, elle assure totalement le verrouillage du couple de torsion. Elle permet outre une correction des diverses déformations et une ré-axation de l'arrière-pied dans les 3 plans de l'espace.

Comparación de perfil lipídico por sexo y edad en una población de equinos en Caldas (Colombia)

Objective: to compare the lipid profile in equines and to analyze the correlation bet-ween serum lipid amounts of four groups. Materials and Methods: Blood samples were obtained from 160 horses in the fasting state, they were differentiated by sex and age without discriminating race as follows: young (40) and adult males (40) and young (40) and adult females (40), the females were not pregnant; the levels of triglycerides (TG), total cholesterol (TC) and high-density lipoprotein cholesterol (HDL-C) were determined by the enzymatic colorimetric method. Very low density (C-VLDL) and low density (LDL-C) lipoprotein cholesterol was determined using the Friedewald formula. The results were compared using a statistical analysis of SIMPLE ANOVA with the statistical program IBM SPSS Statistics 23®, where a statistically significant difference was accepted when P < 0.05. Results: The variables of CT, TG, C-HDL, C-LDL and C-VLDL of the total population according to mean and standard deviation showed values expressed in mg/dl of 100.3 ± 12.89; 45.59 ± 11.95; 62.03 ± 10.88; 27.91 ± 6.48; 9.11 ± 2. When comparing the 4 groups we found a significant difference in HDL-C in adults (p-value: 0.031), being higher in adult females. Conclusions: a statistically significant difference between genotype for HDL-C was found to be higher in adult females, whereas for the rest of the analyzed elements (TG, CT, C-LDL, C-VLDL) no significant difference between gender and age was found.Keywords: lipid,metabolism, equine.
Objetivo: comparar el perfil lipídico en equinos y analizar la correlación entre las cantidades lipídicas del suero de cuatro grupos. Materiales y Métodos: se obtuvieron muestras de sangre de 160 equinos en estado de ayuno, los cuales se diferenciaron por sexo y edad sin discriminar la raza así: machos jóvenes (40) y adultos (40), y hembras jóvenes (40) y adultas (40), las hembras no estaban preñadas; se determinaron los niveles de triglicéridos (TG), colesterol total (CT) y el colesterol de las lipoproteínas de alta densidad (C-HDL) mediante el método enzimático colorimétrico. El colesterol de las lipoproteínas de muy baja densidad (C-VLDL) y de baja densidad (C-LDL) se determinó usando las fórmulas de Friedewald. Los resultados se compararon mediante un análisis estadístico de Anova simple con el programa estadístico IBM SPSS Statistics 23®, donde se aceptaba diferencia estadísticamente significativa cuando p < 0,05. Resultados: las variables de CT, TG, C-HDL, C-LDL y C-VLDL del total de la población de acuerdo a la media y desviación estándar mostraron valores expresados en mg/dl de 100,3 ± 12,89; 45,59 ± 11,95; 62,03 ± 10,88; 27,91 ± 6,48; 9,11 ± 2. Al comparar los 4 grupos se encontró diferencia significativa en el C-HDL en los adultos (p-valor: 0,031), siendo mayor en las hembras adultas. Conclusiones: se presenta una diferencia estadísticamente significativa entre género para C-HDL siendo más alto en hembras adultas, mientras que para el resto de los elementos analizados (TG, CT, C-LDL, C-VLDL) no se encontró diferencia significativa entre género y edad.

Hippolyte Tautain. Origine littéraire et identité sémantique d’un personnage de Madame Bovary

L’identité et la fortune d’Hippolyte, personnage de Madame Bovary, peuvent être interprétées comme une écriture moderne de la fortune de l’Hippolytos mythique des tragédies d’Euripide, de Sénèque et de Racine, qui ont toutes traité de sa fin tragique. Un des choix programmatiques de Flaubert, comme le montre le lemme « Hippolyte » du Dictionnaire des idées reçues, a été de retravailler le récit de l’Hippolytos ancien. Flaubert invente Hippolyte, le transforme en une victime et le torture en faisant de lui un handicapé, pour railler l’absence de tolérance de la société à l’égard du handicap corporel du garçon d’écurie et surtout pour critiquer l’incompétence, la recherche du profit personnel, la vanité et la fatuité des protagonistes du roman. Cette « conjuration » qui conduit Hippolyte à la destruction reproduit dans la ville imaginaire du xixe siècle le scénario de la fortune de l’Hippolytos ancien, selon lequel Aphrodite, Phèdre, Thésée, Poséidon et ses auxiliaires « collaborent » pour faire du mal au jeune homme. Or, l’invention flaubertienne la plus originelle est la création de l’identité sémantique d’Hippolyte : en insérant le nom « Hippolyte » dans le récit, Flaubert remanie les composants du nom grec ancien « Hippolytos », dérivé du substantif ἵππος (cheval) et du verbe λύω (délier, détacher ; détruire) ; à un niveau littéraire ces composants sont associés à la fin tragique du héros (« celui qui a été démembré par ses chevaux »). Les significations du nom évoluent au cours du récit, à partir de sa première apparition comme garçon d’écurie.
