Agrobacterium tumefaciens Tumor Morphology Root Plastid Localization and Preferential Usage of Hydroxylated Prenyl Donor Is Important for Efficient Gall Formation1CWOA
Upon Agrobacterium tumefaciens infection of a host plant, Tumor morphology root (Tmr) a bacterial adenosine phosphate-isopentenyltransferase (IPT), creates a metabolic bypass in the plastid for direct synthesis of trans-zeatin (tZ) with 1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate as the prenyl donor. To understand the biological importance of Tmr function for gall formation, we compared Tmr and Trans-zeatin secretion (Tzs) another agrobacterial IPT that functions within the bacterial cell. Although there is no significant difference in their substrate specificities in vitro, ectopic overexpression of Tzs in Arabidopsis (Arabidopsis thaliana) resulted in the accumulation of comparable amounts of tZ- and N(6)-((2)-isopentenyl)adenine (iP)-type cytokinins, whereas overexpression of Tmr resulted exclusively in the accumulation of tZ-type cytokinins. Ectopic expression of Tzs in plant cells yields only small amounts of the polypeptide in plastid-enriched fractions. Obligatory localization of Tzs into Arabidopsis plastid stroma by translational fusions with ferredoxin transit peptide (TP-Tzs) increased the accumulation of both tZ- and iP-type cytokinins. Replacement of tmr on the Ti plasmid with tzs, TP-tzs, or an Arabidopsis plastidic IPT induced the formation of smaller galls than wild-type A. tumefaciens, and they were accompanied by the accumulation of iP-type cytokinins. Tmr is thus specialized for plastid localization and preferential usage of 1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate in vivo and is important for efficient gall formation.
oxLDL-Induced Lipid Accumulation in Glomerular Podocytes: Role of IFN-gamma, CXCL16, and ADAM10
Previous studies have shown that lipid accumulation plays an important role in the pathogenesis and development of glomerular sclerosis. oxLDL caused damage in renal mesangial cells, endothelial cells, and podocytes, and podocytes might be the major victim of oxLDL insult. However, the regulatory mechanism of how oxLDL induces the damage of podocytes remains to be elucidated. In this study, oil red staining was used to investigate the lipid accumulation in podocytes. Moreover, the effects of CXCL16 antibody, IFN-γ, and ADAM10 inhibitor on oxLDL intake and CXCL16 expression were also explored to elucidate the regulatory factors of lipid accumulation in podocytes.[PUBLICATION ABSTRACT]
Quantifying the compound hazard of freezing rain and wind gusts across CONUS
The co-occurrence of freezing rain, ice accumulation and wind gusts (FZG) poses a significant hazard to infrastructure and transportation. However, quantification of the frequency and intensity of FZG is challenged by the lack of direct icing measurements. In this work, we evaluate and then apply an energy balance model to high-frequency data collected during 2005–2022 to derive hourly ice accumulation at 883 stations across the contiguous USA. These estimates are combined with wind gust observations to compute time series of hourly FZG hazard magnitude using the Sperry–Piltz Ice Accumulation (SPIA) index. Results are evaluated using Storm Reports of damage and economic disruption. The hourly SPIA estimates are also used to (i) derive a geospatial atlas of the hazard including the 50 yr return period event intensities for each US state derived using superstations, and (ii) describe storylines of significant events in terms of meteorological drivers and socioeconomic impacts. The highest values of SPIA during the 18 yr study period occur in a region extending from the Southern Great Plains over the Midwest into the densely populated Northeast. States in these regions also have high 50 yr return period maximum radial ice accumulation of 3–5 cm and co-occurring wind gusts >30 ms −1 . These values are comparable to past estimates for the 500 yr event which may imply this hazard has been previously underestimated. This atlas can be used to inform optimal FZG hazard mitigation strategies for each state/region.
Accumulated water and category of modelling: a case with Chilean student teachers
This research analyses the resignification of the definite integral in a category of modelling that emerged during the development of a specific situation of change, in a physics context, with an accumulation argumentation. A case study was conducted with 12 students of mathematics pedagogy. The resignification was analyzed using the theoretical constructs of the category of modelling and documentary analysis techniques. The results indicated that the students resignified a definite integral with meanings associated with water accumulation. The mathematical knowledge of students that emerged in the situation can be incorporated into school mathematics to contribute to an educational change that confronts and values knowledge of school and non-school mathematical scenarios.
Regional Antarctic snow accumulation over the past 1000 years
Here we present Antarctic snow accumulation variability at the regional scale over the past 1000 years. A total of 79 ice core snow accumulation records were gathered and assigned to seven geographical regions, separating the high-accumulation coastal zones below 2000 m of elevation from the dry central Antarctic Plateau. The regional composites of annual snow accumulation were evaluated against modelled surface mass balance (SMB) from RACMO2.3p2 and precipitation from ERA-Interim reanalysis. With the exception of the Weddell Sea coast, the low-elevation composites capture the regional precipitation and SMB variability as defined by the models. The central Antarctic sites lack coherency and either do not represent regional precipitation or indicate the model inability to capture relevant precipitation processes in the cold, dry central plateau. Our results show that SMB for the total Antarctic Ice Sheet (including ice shelves) has increased at a rate of 7 ± 0.13 Gt decade−1 since 1800 AD, representing a net reduction in sea level of ∼ 0.02 mm decade−1 since 1800 and ∼ 0.04 mm decade−1 since 1900 AD. The largest contribution is from the Antarctic Peninsula (∼ 75 %) where the annual average SMB during the most recent decade (2001–2010) is 123 ± 44 Gt yr−1 higher than the annual average during the first decade of the 19th century. Only four ice core records cover the full 1000 years, and they suggest a decrease in snow accumulation during this period. However, our study emphasizes the importance of low-elevation coastal zones, which have been under-represented in previous investigations of temporal snow accumulation.
Post-stress bacterial cell death mediated by reactive oxygen species
Antimicrobial efficacy, which is central to many aspects of medicine, is being rapidly eroded by bacterial resistance. Since new resistance can be induced by antimicrobial action, highly lethal agents that rapidly reduce bacterial burden during infection should help restrict the emergence of resistance. To improve lethal activity, recent work has focused on toxic reactive oxygen species (ROS) as part of the bactericidal activity of diverse antimicrobials. We report that when Escherichia coli was subjected to antimicrobial stress and the stressor was subsequently removed, both ROS accumulation and cell death continued to occur. Blocking ROS accumulation by exogenous mitigating agents slowed or inhibited poststressor death. Similar results were obtained with a temperature-sensitive mutational inhibition of DNA replication. Thus, bacteria exposed to lethal stressors may not die during treatment, as has long been thought; instead, death can occur after plating on drug-free agar due to poststress ROS-mediated toxicity. Examples are described in which (i) primary stress-mediated damage was insufficient to kill bacteria due to repair; (ii) ROS over-came repair (i.e., protection from anti-ROS agents was reduced by repair deficiencies); and (iii) killing was reduced by anti-oxidative stress genes acting before stress exposure. Enzymatic suppression of poststress ROS-mediated lethality by exogenous catalase supports a causal rather than a coincidental role for ROS in stress-mediated lethality, thereby countering challenges to ROS involvement in anti-microbial killing. We conclude that for a variety of stressors, lethal action derives, at least in part, from stimulation of a self-amplifying accumulation of ROS that overwhelms the repair of primary damage.
An apple MYB transcription factor regulates cold tolerance and anthocyanin accumulation and undergoes MIEL1‐mediated degradation
Summary MYB transcription factors (TFs) have been demonstrated to play diverse roles in plant growth and development through interaction with basic helix‐loop‐helix (bHLH) TFs. MdbHLH33, an apple bHLH TF, has been identified as a positive regulator in cold tolerance and anthocyanin accumulation by activating the expressions of MdCBF2 and MdDFR. In the present study, a MYB TF MdMYB308L was found to also positively regulate cold tolerance and anthocyanin accumulation in apple. We found that MdMYB308L interacted with MdbHLH33 and enhanced its binding to the promoters of MdCBF2 and MdDFR. In addition, an apple RING E3 ubiquitin ligase MYB30‐INTERACTING E3 LIGASE 1 (MdMIEL1) was identified to be an MdMYB308L‐interacting protein and promoted the ubiquitination degradation of MdMYB308L, thus negatively regulated cold tolerance and anthocyanin accumulation in apple. These results suggest that MdMYB308L acts as a positive regulator in cold tolerance and anthocyanin accumulation in apple by interacting with MdbHLH33 and undergoes MdMIEL1‐mediated protein degradation. The dynamic change in MYB‐bHLH protein complex seems to play a key role in the regulation of plant growth and development.
Cascading trend of Early Paleozoic marine radiations paused by Late Ordovician extinctions
The greatest relative changes in marine biodiversity accumulation occurred during the Early Paleozoic. The precision of temporal constraints on these changes is crude, hampering our understanding of their timing, duration, and links to causal mechanisms. We match fossil occurrence data to their lithostratigraphical ranges in the Paleobiology Database and correlate this inferred taxon range to a constructed set of biostratigraphically defined high-resolution time slices. In addition, we apply capture–recapture modeling approaches to calculate a biodiversity curve that also considers taphonomy and sampling biases with four times better resolution of previous estimates. Our method reveals a stepwise biodiversity increase with distinct Cambrian and Ordovician radiation events that are clearly separated by a 50-million-year-long period of slow biodiversity accumulation. The Ordovician Radiation is confined to a 15-million-year phase after which the Late Ordovician extinctions lowered generic richness and further delayed a biodiversity rebound by at least 35 million years. Based on a first-differences approach on potential abiotic drivers controlling richness, we find an overall correlation with oxygen levels, with temperature also exhibiting a coordinated trend once equatorial sea surface temperatures fell to present-day levels during the Middle Ordovician Darriwilian Age. Contrary to the traditional view of the Late Ordovician extinctions, our study suggests a protracted crisis interval linked to intense volcanism during themiddle Late Ordovician Katian Age. As richness levels did not return to prior levels during the Silurian—a time of continental amalgamation—we further argue that plate tectonics exerted an overarching control on biodiversity accumulation.
