Overview
In a phase 2, placebo-controlled trial, nemolizumab, an antibody against interleukin-31 receptor A, reduced pruritus in patients with moderate-to-severe atopic dermatitis. These findings support the role of interleukin-31 in the pathophysiology of atopic dermatitis.
Atopic dermatitis is a chronic pruritic, inflammatory skin disease that is triggered by an immune response to antigenic substances, irritants, and mechanical irritation and which is often associated with a personal or family history of type 1 allergies, allergic rhinitis, or asthma.

Pruritus often causes patients to persistently scratch their skin, leading to sleep disturbance and exacerbation of atopic dermatitis. Pruritus has a negative effect on the patients’ quality of life, with the intensity of pruritus directly affecting psychosocial well-being.
