Overview

This phase 2 trial investigated the efficacy of motesanib diphosphate, an orally active inhibitor of VEGF receptors, in patients with advanced differentiated thyroid cancer. None of the patients had had a response to conventional treatment. No patient had a complete response, but 14% had a partial response, with a median duration of 32 weeks. Motesanib diphosphate is an option for patients with metastatic or locally progressive differentiated thyroid cancer who have not had a response to usual treatment. This trial investigated the efficacy of motesanib diphosphate in patients with advanced differentiated thyroid cancer. No patient had a complete response, but 14% had a partial response, with a median duration of 32 weeks. Thyroid cancers are classified histologically into four groups: papillary, follicular, medullary, and undifferentiated or anaplastic thyroid carcinomas. 1 Papillary and follicular carcinomas (including the Hürthle-cell variant) are collectively known as differentiated thyroid cancers, 1 , 2 and they account for approximately 95% of incident cases. 3 Initial treatment typically involves total thyroidectomy followed by long-term administration of levothyroxine at doses that suppress thyrotropin; adjuvant radioiodine ( 131 I) is often administered as well. 2 – 4 This regimen is usually effective, but the 10-year recurrence rate is 20 to 30% among patients who are older, among patients with tumors larger than 4 cm in diameter, and among . . .
